Three‐dimensional mass spectral imaging of HeLa‐M cells – sample preparation, data interpretation and visualisation

Abstract: Time-of-flight secondary ion mass spectrometry (ToFSIMS) is being applied increasingly to the study of biological systems where the chemical specificity of mass spectrometry and the high lateral resolution imaging capabilities can be exploited. Here we report a comparison of two cell sample preparation methods and demonstrate how they influence the outcome of the ToFSIMS analysis for three-dimensional (3D) imaging of biological cells using our novel buncher-ToF instrument (J105 3D Chemical Imager) equipped wit… Show more

“…Care must also be taken when mounting/preparing samples to ensure no height profile artifacts are introduced. Additional care must be taken when mounting cells for analysis in order to avoid analyte delocalization [154]. Methods for the preparation of biological samples for SIMS have been covered in a recent review by Passareli and Winograd and usually involve cryofixation, sectioning and freeze drying for tissue and frozen hydration for cells [21].…”

“…Nonetheless, SIMS is currently the only MS-based method providing sub-micron spatial resolution and allowing the distribution of lipids across a cell surface, including 3-D lipid distributions, to be investigated [154], and thus represents an important tool for lipid analysis.…”

“…[5,12] Successful attempts to increase secondary ion yields involved the enhancement of the protonation of ions with the presence of water during the analysis. [6][7][8] There have been attempts to analyse the sample in a frozen hydrated state (instead of freeze-drying the sample) [9][10][11]13] or to inject water vapour at the sample's surface. [14] Another approach proposed the use of a water vapour electrospray beam as the primary ion beam to explore its effect on secondary ion yields.…”

“…The J105 instrument is an exception to this rule as it uses a DC primary ion beam and therefore is capable of collecting high spatial resolution images with good mass resolution. 27,28) Even with this advantage, the J105 is still limited by low ion yields from biological materials.…”
