Therapeutic effects of <i>Ligularia stenocephala</i> against inflammatory bowel disease by regulating antioxidant and inflammatory mediators

The aim of this work was to evaluate the potential anti-inflammatory effect of protein hydrolysate and peptide fractions from Salvia hispanica L. seeds. Protein isolate was obtained using defatted flour of mucilage-free seeds. Hydrolysis process was conducted by enzymatic digestion. The hydrolysate was fractionated using ultrafiltration membranes to obtain the peptide fractions (<1, 1-3, 3-5, 5-10, and >10 kDa). Protein derivatives were evaluated by in vitro activation of murine peritoneal macrophages. The antiinflammatory activity was determined as NO production, H 2 O 2 release and pro-and anti-inflammatory cytokines (TNF-α, IL-1β, IL-6, and IL-10) production. All the peptides exerted an antiinflammatory activity, but peptide fraction between 1-3 kDa showed the highest anti-inflammatory effect. This fraction was evaluated on in vivo murine models of TPA-induced ear edema and DNFB-induced delayed-type hypersensitivity, exhibiting inhibitory effects. Hence, the results demonstrated that protein derivatives from S. hispanica L. seeds have in vitro and in vivo antiinflammatory effects.

Bilberry anthocyanin-rich extract protects against retinal photooxidative damage via activation of HO-1 and inhibition of NF-κB

Bilberry anthocyanins are functional ingredients for preventing retinal degeneration. This study was to determine whether the molecular mechanism of bilberry anthocyanin-rich extract (BAE) protection of retinal pigment epithelial (RPE) cells and retina against photooxidative damage via heme oxygenase-1 (HO-1) upregulation in vitro and in vivo. Results shown that BAE protected ARPE-19 cells from visible light-induced damage, reduced the intracellular reactive oxygen species levels, and up-regulated the expression of HO-1. Pigmented rabbits were treated with BAE (250 and 500 mg/kg/day) for 2 weeks pre-illumination and 1 week postillumination until sacrifice. The expression of HO-1 in the retina was up-regulated after BAE treatment. BAE significantly suppressed the photooxidation-induced increase in expression of interleukine-6 protein. BAE also attenuated the expression of inflammationrelated gene nuclear factor kappa B (NF-κB). Thus, BAE reduced retinal photooxidative damage through activating the HO-1 expression and suppressing NF-κB activation. ARTICLE HISTORY

Oat (Avena sativa) bran, a by-product during oat grain processing, is a excellent source of fibre, vitamins and phytochemicals. The health benefits of dietary fibre have long been established. In the present study, the antioxidant and hepatoprotective effects of oat bran extracts were investigated. The extracts showed strong 1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2 ′ -azino-bis(3-ethylbenzothiazoline-6sulfonic acid) diammonium salt (ABTS), hydroxyl, superoxide radicals scavenging and reducing power activities in vitro. In addition, the extracts (50 mg/kg mouse) significantly reduced the levels of malondialdehyde and hepatic damage marker enzymes (aspartate transaminase and alanine transaminase), proinflammatory cytokines as well as elevated SOD, CAT, and GPx enzymes expressions in LPStreated (1 mg/kg body weight) BALB/c mice. Therefore, the present study strongly suggests that both oat bran extracts could be excellent raw materials for manufacturing functional food due to its protective effect on hepatic liver damage and excellent antioxidant activity.
