Therapeutic effect of simvastatin on DMBA‐induced breast cancer in mice

Abstract: Preclinical studies have shown positive effects of statins against specific cancers. This study aimed to determine the therapeutic effect of simvastatin in 12-dimethylbenz(a)anthracene (DMBA)-induced breast cancer. Female albino mice were divided into two groups, with or without DMBA administration. After tumor appearance, DMBA-treated group was further divided into four groups (D1-D4) as control (D1), treated with simvastatin at 80 and 40 mg/kg/day, orally (D2 and D3) and tamoxifen (50 mg/kg/day, orally) trea… Show more

“…It is recorded that the tumor size significantly decreased after 16 days of treatment (707 mm³ vs. 825 mm³ in control, Pvalue=0.01) compared to the control. These findings approved antitumor effect of simvastatin on solid tumor animal model and agreed with the finding on a rat model of malignant glioma [26] and breast cancer in the mice [27]. Simvastatin recorded good results of ILS% (31.58), T/C (132) and TIR% is 14.28%.…”

“…DMBA promotes the production of prostaglandin E2 (PGE2) which in turn increase the incidence of mammary tumors . In addition to other carcinogenic properties as mutation and disturbance of cellular oxidant-antioxidant balance (9).…”

“…The mice were weighed and palpated weekly for the appearance of mammary tumors 4 weeks after DMBA administration till to the end of the experiment. When tumors size reached 0.5cm in the largest dimension, animals in group D were divided into three groups, 7 in each group (Karimi et al, 2018). Groups were labeled as follows: C, tumoric mice without treatment which were represented as the control group (n=7); S, tumeric mice with simvastatin treatment that received it by gavage with a dose of 80 mg/kg/day for 4 consecutive weeks (n=7) and T: tumeric mice with tamoxifen treatment that received it by gavage with a dose of 50 mg/kg/day for 4 consecutive weeks (n=7).…”
