Background: Cyclophosphamide (CP) is an anticancer drug utilized for treatment of many types of cancer. CP causes many side effects such as hepatotoxicity and mutagenicity. Garlic has long been used as a spice and food additive and is considered as one of the important medicinal plants. Garlic had medicinal importance as it has hepatoprotective, antimicrobial, antioxidant, anti-inflammatory, antithrombotic, antihypertensive, and antihyperlipidemic properties. The purpose of the presented study is to investigate the protective role of aged black garlic against cyclophosphamide-induced hepatotoxicity and apoptosis. The present work was carried out by routine histological H&E stain, histochemical Feulgen stain for DNA, immunohistochemical stain Bcl-2 as apoptotic marker, comet assay, serum biochemical analysis for liver function markers (ALT, AST, albumin, and bilirubin), and hepatic oxidative stress markers (MDA, CAT, SOD, and GPx). Results: Cyclophosphamide induced histological alteration as fatty degeneration, blood venous congestion, pyknosis, necrosis, and vaculations. CP caused a decrease of DNA content, an increase of Bcl-2 immunoreactivity, and increase of DNA damage. Biochemical investigations showed that CP induced significant elevation of ALT, AST, bilirubin, and hepatic MDA. Moreover, CP induced reduction of albumin and hepatic SOD, CAT, and GPx values. Aged black garlic induced significant improvement of alterations induced by CP, when compared with CP-treated group. Conclusion: Aged black garlic succeeded in protecting the liver from apoptosis and pathological alterations induced by CP.

Kushen (Radix Sophorae Flavescentis) has a long history of use for the treatment of tumors, inflammation and other diseases in traditional Chinese medicine. Compound Kushen Injection (CKI) is a mixture of natural compounds extracted from Kushen and Baituling (Rhizoma Smilacis Glabrae). The main principles of CKI are matrine (MT) and oxymatrine (OMT) that exhibit a variety of pharmacological activities, including anti-inflammatory, anti-allergic, anti-viral, anti-fibrotic and cardiovascular protective effects. Recent evidence shows that these compounds also produce anti-cancer actions, such as inhibiting cancer cell proliferation, inducing cell cycle arrest, accelerating apoptosis, restraining angiogenesis, inducing cell differentiation, inhibiting cancer metastasis and invasion, reversing multidrug resistance, and preventing or reducing chemotherapy-and/or radiotherapy-induced toxicity when combined with chemotherapeutic drugs. In this review, we summarize recent progress in studying the anti-cancer activities of MT, OMT and CKI and their potential molecular targets, which provide clues and references for further study.

Aim: The present study aimed to investigate the protective effect of Artemisia Judaica (A. Judaica) against doxorubicin (DOX)-induced toxicity in male mice. Place and Duration of Study: Department of Cell Biology, Genetic engineering and biotechnology division, National research centre, Egypt, between March 2016 and February 2017. Methodology: Male mice were divided into 7 groups (n=10) and treated as follow: the control group, the group treated with DMSO, the group injected (i.p.) with DOX , the groups treated with low and high dose of A. judaica extract and the groups injected (i.p.) with DOX and treated with low and high dose of A. judaica extract. Femur, testes and liver samples were collected for different analyses. Results: Our data showed that A. judaica significantly reversed the DOX-induced elevation of DNA fragmentation rate and MDA level in liver tissue, as well as declined chromosomal aberrations (CAs) either in the bone marrow cells or in the spermatocyte cells. Meanwhile, the expression of Ahmed et al.; ARRB, 18(3): 1-10, 2017; Article no.ARRB.35990 2 apoptosis-related genes (Bax and Caspase-3) in liver tissues was analyzed by quantitative real-time PCR (qRT-PCR) and results revealed that genes expression were up-regulated in DOX treated mice however; the administration of A. judaica didn't alter such increase. Conclusion: Overall, the findings indicated that A. judaica may attenuate the DOX-induced toxicity. However; further studies are required to confirm the protective effect of A. judaica extract against toxicity caused by DOX drug. Original Research Article
