In this study, we analyzed the virome of 73 grape samples from two Dagestan ampelographic collections in Russia using high-throughput sequencing of total RNAs. Fourteen viruses and four viroids were identified, with one to eleven of them detected in each plant. For the first time in Russia, we identified grapevine leafroll-associated virus 7 and grapevine Kizil Sapak virus. A total of 206 genomes of viruses and viroids were obtained, and their phylogenetic analysis was carried out. The de novo assembly and tblastx analysis allowed us to obtain contigs of a novel (+) ssRNA genome of a plant virus from the genus Umbravirus, which was tentatively named grapevine umbra-like virus (GULV), as well as contigs of a novel dsDNA pararetrovirus from the genus Caulimovirus, which was tentatively named grapevine pararetrovirus (GPRV). Complete genomes of these viruses were obtained and used for Sequence Demarcation Tool (SDT) analysis and phylogeny studies. GULV and GPRV were detected in 16 and 33 germplasm samples from the Dagestan collections, respectively.

As the sensitivity of perennial crops to climate change becomes more pronounced, clonal selection, which is already very time-consuming for grapevine, may take even longer, while its importance is increasing. In the case of indigenous grapevine varieties, the purpose of clonal selection is twofold: to mitigate problems of cultivation and at the same time, to preserve the varietal character. The cultivation technique issue of ‘Kéknyelű’ is the low fertility (functionally female-flowered variety), and as for ‘Juhfark’ it is the significant susceptibility to grey rot. Based on daily meteorological data of 11 years, the years were classified into 3 groups and harvest data were analyzed within each group. Significant difference in yield was found between clone B.2. and the base ‘Kéknyelű’. Both clones of ‘Kéknyelű’ matured with significantly lower pH compared to the base variety. Given the acidic character of ‘Kéknyelű’ wine and the predicted rise in must °Brix and pH as a result of climate change, these differences may be useful in the future. Botrytis infection only showed statistically significant differences between year groups for ‘Juhfark’. It is intriguing that in most years, the rate of grey rot infection was lower in both clones compared to the base variety, especially in year groups 1 and 3 when the overall rate of Botrytis infection was quite low.

The share of seedless grape varieties in the total grape production in the country is very small. Of the 106 table varieties included in the register, only 4 are seedless grape varieties (Kishmish Luchistiy, Korinka Russkaya, Yuzhnoberezhniy, Yaltinskiy Bessemyanniy). These varieties are very much in demand, as their berries are used both fresh and dried. This article shows the results of multi-year research on the identification of seedless genetic donors among grape varieties of the selection of the Anapa Zonal Experimental Station for Grape Growing and Wine Making, such as Lotos, Zhemchug Anapy and Kishmish Rozovy AZOS. In the frame of this study, agronomic characters and annually changing weather conditions were studied side by side. In addition, an assessment of these varieties was carried out in terms of the intensity of the seedless character in the conditions of the Anapa-Taman grape growing zone. The largest mass of rudiments in all the analyzed samples was found in the Kishmish Rozovy AZOS variety; therefore, it was assigned to the third category of seedless grape, although the ratio of the rudiment mass to the berry mass is negligible. The rest of the studied seedless grape varieties in comparison with the control variety (Kishmish Luchistiy) have a higher seedless class. In particular, this concerns the varieties – Zhemchug Anapy and Lotos belonging to the first and second seedless class, respectively. At the same time, yield of these varieties and their productivity (fruit-bearing and fruitfulness coefficients) together with plant development phenology depending on weather conditions were studied for several years. In particular, it was found that annually high productivity indicators are observed in all the studied varieties, as well as high yields are observed every year in the varieties Lotos and Zhemchug Anapy. This article presents the results of these studies from 2017 to 2019. The work performed showed the prospect of using productive, resistant to biotic and abiotic environmental factors with a high seedless category of grape varieties of the AZESGGaWM selection as future seedless donors.
