Flax (Linum usitatissimum L.) products are used in the food, pharmaceutical, textile, polymer, medical, and other industries. The creation of a pan-genome will be an important advance in flax research and breeding. The selection of flax genotypes that sufficiently cover the species diversity is a crucial step for the pan-genomic study. For this purpose, we have adapted a method based on Illumina sequencing of transcriptome libraries prepared using the Tn5 transposase (tagmentase). This approach reduces the cost of sample preparation compared to commercial kits and allows the generation of a large number of cDNA libraries in a short time. RNA-seq data were obtained for 192 flax plants (3–6 individual plants from 44 flax accessions of different morphology and geographical origin). Evaluation of the genetic relationship between flax plants based on the sequencing data revealed incorrect species identification for five accessions. Therefore, these accessions were excluded from the sample set for the pan-genomic study. For the remaining samples, typical genotypes were selected to provide the most comprehensive genetic diversity of flax for pan-genome construction. Thus, high-throughput sequencing of tagmentation-based transcriptome libraries showed high efficiency in assessing the genetic relationship of flax samples and allowed us to select genotypes for the flax pan-genomic analysis.

The article is devoted to the systematization of scientific developments of domestic scientists in the sphere of oilseeds flax processing and their use in technologies of flour products. Oilseed flax is the only oilseed crop in which almost all anatomical parts are important for industrial processing. The use of oil flax in the food industry is due not only to the presence of biologically valuable nutrients, but also to the functional and technological properties inherent in the products of their processing. It is shown that along with whole and ground flax seeds, not only flax seeds processing products (full-fat, semi-fattened, skimmed and oil), but also bran and cryopowder from seeds are widely used in the above food technologies. Flax seed hydrocolloids or dry polysaccharide extract deserve special attention, which are of interest not only from the technological point of view (analog of xanthan gum, guar gum), but also from the medical and biological position, since they perform prebiotic function in the human body. Analysis of numerous domestic sources of scientific, technical and patent information allows us to conclude that flax seeds are, on the one hand, a well-known and widely used raw material. On the other hand, it should be noted that there are still a number of insufficiently elaborated directions, in particular, additional research is needed, which will allow to realize on an industrial scale economically competitive technologies of production of flour products, maximizing the potential (technological and medical and biological) of non-starch polysaccharides, lignans and other biologically active compounds of flax seeds

Genotyping oil flax varieties using the microsatellite DNA marker system

The tendency to increase crop acreage of oil flax requires the development of new varieties with high indicators of ecological plasticity, productivity and resistance to biotic and abiotic stresses. The application of modern biotechnolog ical approaches based on the use of molecular markers can accelerate the assessment of genetic differences and the dete rmination of potential of the source material for breeding. The research was aimed at assessment of the genotyping parameters of some oil flax varieties of VNIIMK breeding using the system of microsatellite markers. Seventeen variety samples of flax were used as the material for the research. DNA was isolated using CTAB buffer. Eleven SSR loci were used for the identification of varieties. Ten polymorphic loci were identified during the research. The total number of counted alleles is 32. The size of alleles varied in the range of 111-210 bps. The number of alleles per locus ranged from 2 to 6 with an average value of 3.20. The value of the index of polymorphic information content (PIC) was from 0.29 to 0.75 with an average parameter value of 0.51. The effective number of alleles for different loci is determined in the range of 1.40-3.94 with an average value of 2.28. The level of information content of the marker system (PIC 0.51) corresponds to that for identifying sets of genotypes from collections with a limitation in geographical origin. There were established differences in the frequency of occurrence of alleles. The discriminatory potential of the used marker system allowed to identify 15 variety samples. Two genotypes with common origin were identical. The degree of genetic relatedness of the studied flax genotypes has been evaluated. The obtained results will serve as the basis for the subsequent construction of genetic passports of oil flax varieties of VNIIMK breeding.
