In this study, Arthrospira fusiformis previously isolated from Lake Mariout (Alexandria, Egypt) was cultivated in the laboratory using a medium for pharmaceutical grade Arthrospira, named as Amara and Steinbüchel medium. Hot water extract of the Egyptian Spirulina was prepared by autoclaving dried biomass in distilled water at 121°C for 15 min. This algal water extract was analyzed by GC-MS to evaluate its volatile compounds and fatty acids composition. The antimicrobial activity of phycobiliprotein extract from Arthrospira fusiformis using phosphate buffer was evaluated against thirteen microbial strains (two Gram-positive bacteria, eight Gram-negative bacteria, one yeast, and two filamentous fungi). The major components of fatty acids in the hot extract of Egyptian A. fusiformis were hexadecanoic acid (palmitic acid, 55.19%) and octadecanoic acid (stearic acid, 27.14%). The main constituents of its volatile compounds were acetic acid (43.33%) and oxalic acid (47.98%). The most potent antimicrobial effect of phycobiliprotein extract was obtained against two Gram-negative bacteria Salmonella typhi and Proteus vulgaris, filamentous fungus Aspergillus niger, and the pathogenic yeast Candida albicans (all of which showed MIC values of 58.1 μg/ml). Escherichia coli and Salmonella typhimurium come second in their susceptibility to the phycobiliprotein extract from Arthrospira fusiformis and Serratia marcescens and Aspergillus flavus are the least in susceptibility, with MIC values of 116.2 and 232.5 μg/ml, respectively, while phycobiliprotein extract has no antibacterial effect on methicillin-resistant as well as susceptible Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Shigella sonnei. These findings confirmed the nutritional value of Egyptian A. fusiformis isolated from Lake Mariout and suggest the potential use of this strain as an ingredient in the cooking of some foods to increase the level of stearic acid and palmitic acid. Moreover, its effective antibacterial activities against some important and highly resistant to antibiotics bacterial pathogens in addition to its antifungal effects recommend the therapeutic use of its biomass.

Potential Activity of Arthrospira platensis as Antioxidant, Cytotoxic and Antifungal against Some Skin Diseases: Topical Cream Application

This research evaluated the antifungal effectiveness of Arthrospira platensis ethanol, methanol, ethyl acetate and acetone extracts against the tested pathogenic fungi (Candida albicans, Trichophyton rubrum and Malassezia furfur). Antioxidant and cytotoxicity effectiveness of A. platensis extracts against four distinct cell lines were also assessed. Methanol extract of A. platensis exhibited the highest inhibition zones against Candida albicans as measured by the well diffusion method. A transmission electron micrograph of the treated group of Candida cells with A. platensis methanolic extract showed mild lysis and vacuolation of the cytoplasmic organelles. In vivo, after induced infection of mice by C. albicans and treatment with A. platensis methanolic extract cream, the skin layer emerged with the removal of Candida spherical plastopores. The extract of A. platensis recorded the highest antioxidant activity using the DPPH (2, 2- diphenyl-1-picrylhydrazyl) scavenging method (IC50 28 mg/mL). A cytotoxicity test using a MTT assay showed that the A. platensis extract had strong cytotoxic activity against the HepG2 cell line (IC50 20.56 ± 1.7 μg/mL) and moderate cytotoxic activity against MCF7 and the Hela cell (IC50 27.99 ± 2.1 μg/mL). Gas Chromatography/Mass Spectroscopy (GC/MS) results revealed that the effective activity of A. platensis extract could be linked to a synergistic impact between their prominent composition as alkaloids, phytol, fatty acids hydrocarbons, phenolics and phthalates. A. platensis extract contains active metabolites that constitute a promising source of antifungal, antioxidant and anti-proliferative compounds for the pharmaceutical drug industry.

Spirulinais an edible cyanobacterium known worldwide for its high nutritional value, and the interest in its biological activity and bioactive compounds, such as pigments, phenolics, peptides, and lipids, has been increasing. Due to the variety of its properties, the potential application of Spirulina in health is wide, including dermatological area. In a context of increasing demand for natural products and actives in cosmetics, this microorganism becomes a great source. This article aims to review what has already been reported in literature about the potential effects of Spirulina or its isolated compounds in skin, for either aesthetic or clinical purposes. In many studies, Spirulina and its components show a good influence in proliferation of dermal fibroblasts and keratinocytes, extracellular matrix, and collagen production, as well as exerting antioxidant and anti‐inflammatory action. Thus, they promote a healthy environment for skin's cells and structure, cooperating for the highlighted anti‐aging, photoprotection, and wound‐healing effects. Some compounds of the cyanobacterium also exert a lighting property through tyrosinase inhibition. Its antimicrobial action can also be advantageous to skin contributing to anti‐acne, antibiofilm, and anti‐herpes effects. In face of many attributes and due to its rich composition, Spirulina presents multi‐benefits and shows an improvement in the general aspect of skin. However, some applications are still in need of studying and more clinical evidence is necessary.
