Stress is an important factor in the etiology of some illnesses such as gastric ulcers and depression. Castilleja tenuiflora Benth. (Orobanchaceae) is used in Mexican traditional medicine for the treatment of gastrointestinal diseases and nervous disorders. Previous studies indicated that organic extracts from C. tenuiflora had gastroprotective effects and antidepressant activity. In this study, we aimed to evaluate the gastroprotective and antidepressant activity of fractions and isolated compounds from the methanolic extract (MECt) of C. tenuiflora in stressed mice. Chromatographic fractionation of MECt produced four fractions (FCt-1, FCt-2, CFt-3, and FCt-4) as well as four bioactive compounds which were identified using TLC, HPLC and NMR analyses. The cold restraint stress (CRS)-induced gastric ulcer model followed by the tail suspension test and the forced swim test were used to evaluate the gastroprotective effect and antidepressant activity of the extract fractions. FCt-2 and FCt-3 at 100 mg/kg had significant gastroprotective and antidepressant effects. All isolated compounds (verbascoside, teniufloroside and mixture geniposide/ musseanoside) displayed gastroprotective effects and antidepressant activity at 1 or 2 mg/kg. The above results allow us to conclude that these polyphenols and iridoids from C. tenuiflora are responsible for the gastroprotective and antidepressant effects.

Background: Lead (Pb) is a toxic environmental pollutant and can exerts toxicity in kidneys. It is known that selenium (Se) has an antagonistic effect on Pb poisoning. However, biological events during the process were not well understood in chicken kidneys.Methods: One hundred and eighty male Hyline chickens (7-day-old) were randomly divided into the control group (offering standard diet and potable water), the Se group (offering Na2SeO3-added standard diet and potable water), the Pb group (offering standard diet and (CH3OO)2Pb-added potable water), and the Pb+Se group (offering Na2SeO3-added standard diet and (CH3OO)2Pb-added potable water). On 30th, 60th, and 90th days, kidneys were removed to perform the studies of histological structure, oxidative stress indicators, cytokines, heat shock proteins, and autophagy in the chicken kidneys.Results: The experimental results indicated that Pb poisoning changed renal histological structure; decreased catalase, glutathione-s-transferase, and total antioxidative capacity activities; increased hydrogen peroxide content; induced mRNA and protein expression of heat shock proteins; inhibited interleukin (IL)-2 mRNA expression, and induced IL-4 and IL-12β mRNA expression; inhibited mammalian target of rapamycin mRNA and protein expression, and induced autophagy-related gene mRNA and protein expression in the chicken kidneys. Supplement of Se mitigated the above changes caused by Pb.Conclusion: Our research strengthens the evidence that Pb induced oxidative stress, inflammation, heat shock response, and autophagy and Se administration alleviated Pb poisoning through mitigating oxidative stress in the chicken kidneys.

Introduction: Some studies are in favor of protective and anti-inflammatory effects of Ocimum basilicum (OB) against renal failure. Objectives: The aim of this study was to explore the protective effects of OB extract against gentamicin (GM)-induced nephrotoxicity in male rat models of acute kidney injury. Materials and Methods: Twenty-seven adult male Wistar rats with the weight of 200-250 g were prepared under standard condition (12 h light/dark cycle, 20 to 22°C temperature). In the gentamicin group (GM), the rats were administrated normal saline (5 mL/kg; oral) for four consecutive weeks and then GM (80 mg/kg/d; intraperitoneal) for another one week. In the third (OB-200+ GM) and fourth (OB-800+ GM) groups, the rats were initially treated with OB extract (200 mg/kg and 800 mg/kg respectively; oral) for four consecutive weeks followed by intraperitoneal administration of 80 mg/kg/d GM for another one week. The control group was treated with normal saline (5 mL/kg; oral) for five consecutive weeks. They were sacrificed under deep anesthesia, then plasma and tissue samples were obtained. Results: Treatment with OB hydro-alcoholic extract decreased BUN, serum Cr, FeNa and the levels of TNF-a and NF-kB and also diminished the rate of apoptosis in renal tubular cells compared to gentamicin group. Conclusion: Our findings provide comprehensive evidence supporting the beneficial role of OB, a traditional medicinal herb, as a potential therapeutic supplement to prevent the incidence and progression of renal impairment in GM-induced renal toxicity.
