The complete mitogenome of the subsocial wood-eating cockroach Cryptocercus kyebangensis was sequenced and compared with that of another Asian cockroach, Cryptocercus relictus and the primitive termite Mastotermes darwiniensis. The complete mitogenome of C. kyebangensis is a closed circular molecule of 15,720 bp, containing13 protein-coding genes, two ribosomal RNA genes (12S rRNA and 16S rRNA), 22 transfer RNA genes, and one D-loop region. Nucleotide composition of the C. kyebangensis mitogenome shows an AT bias (74.4%), in particular, 46.0% A, 28.4% T, 9.7% G, and 15.9% C. GC skews are similar among the mitogenomes of C. kyebangensis, C. relictus, and M. darwiniensis, but AT skew is stronger in Cryptocercus mitogenomes than in the M. darwiniensis mitogenome. The whole mitogenome of C. kyebangensis showed 88.8% nucleotide similarity to that of C. relictus but only 74.9% similarity to that of M. darwiniensis. Phylogenetic analyses, based on amino acid sequences of the 13 mitochondrial proteins, showed that the Cryptocercus clade has a sister relation with termites, including the primitive termite M. darwiniensis. Sister relations of Blattidae and Blattellidae, with the clade Cryptocercidae + Isoptera as a sister group, are also evident. Polyphagidae (Eupolyphaga sinensis) appears to be most basal in the phylogenetic trees, showing sister relations with the remaining cockroaches.

The gut of wood-feeding insects is a microhabitat for a specialized community of microbes, including bacteria and several groups of eukaryotes such as nematodes, parabasalids and fungi. The characterization of gut yeast communities from a variety of insects has shown that certain yeasts often are associated with the insects. The gut of woodfeeding insects is rich in ascomycete yeasts and in particular xylose-fermenting (X-F) and assimilating yeasts have been consistently present in the gut of lignicolous insects. The objective of this study was the characterization of the yeast flora from the gut of the wood roach Cryptocercus sp. (Blattodea: Cryptocercidae). Five wood roaches were collected along the Appalachian Trail near the border between Tennessee and North Carolina, USA. We isolated 18 yeast strains from the wood roaches identified as Sugiyamaella paludigena and Sugiyamaella lignohabitans, xylose-assimilating yeasts, and Scheffersomyces cryptocercus (NRRL Y-48824 T 5 CBS 12658) a new species of X-F yeast. The presence of X-F and certain non X-F yeasts in the gut of the subsocial wood roach Cryptocercus sp. extends the previous findings of associations between certain ascomycete yeasts and lignicolous insects. New combinations were made for 13 asexual members of the Sugiyamaella clade.

In the present "tree-thinking" period, relying on accurate phylogenetic hypotheses is of paramount importance for biologists interested in an evolutionary perspective. In the Blaberidae cockroaches, a well-defined monophyletic family comprising several model species, no such phylogenetic tree is available despite several earlier contributions. Here, using six molecular markers (12S, 16S, 18S, 28S, COI and COII), we investigate the relationships of Blaberidae and compare our results with the traditional morphology-based classification. This resulted in a broad spectrum of situations, from congruent and well-supported hypotheses (e.g., the monophyly of Blaberidae, Oxyhaloinae and (Geoscapheiinae + Panesthiinae)) to incongruent and weakly supported results (e.g., polyphyly of Perisphaerinae). We emphasize that interesting and contrasted situations lie between the two extremities of this spectrum, especially concerning the genera Thanatophyllum Grandcolas, 1991, Phoetalia Stål, 1874, Laxta Walker, 1868 and Pronauphoeta Shelford, 1909. We also discuss the phylogenetic position of two incertae sedis genera (Eustegasta Gerstaecker, 1883 and Gynopeltis Gerstaecker, 1869). We conclude that in-depth signal analyses should be performed to better understand molecular evolution and its consequence on tree reconstruction for this group. As for phylogenetic relationships per se, new markers should be searched for, especially to decipher deeper relationships in Blaberidae.
