Azoreductases are being extensively investigated for their ability to initiate degradation of recalcitrant azo dyes through reduction of azo bonds. There is great interest in studying their diversity, structure, and function to facilitate better understanding and effective application. Current study reports azoreductase enzyme from Bacillus velezensis, which showed 69.5% identity to the Bacillus subtilis azoreductase YhdA. The enzyme was homotetrameric and molecular weight of each subunit was 20 kDa. It decolorized azo dyes with different structures. The Vmax for decolorization of congo red, methyl orange and methyl red was 14.7, 28.6, and 77.9 nmol/min/mg, respectively. The enzyme contained FMN as cofactor and used NADPH as the favored co‐substrate. It was oxygen‐insensitive, but the presence of reducing agents enhanced its activity, which is a new finding. The azoreductase expression in B. velezensis was found to be unaffected by addition of azo dyes, although azo dyes are known to induce azoreductase expression in few organisms. The enzyme was thermostable with melting temperature of 89.5°C and functioned in wide temperature range. Further, the enzyme was crystallized and its structure was solved. The structural basis of its functional attributes is discussed. In our knowledge, this is the first report on characterization of azoreductase enzyme from B. velezensis.

T his work investigated the photodegradation of azo dye, methyl orange, without and with the addition of C-N-codoped TiO 2 catalyst using a visible halogen-lamp as a light source. C-Ncodoped TiO 2 was prepared by free-organic solvent peroxo sol-gel method. The effects of initial dye concentration, catalyst dosage, and pH solution on the photodegradation of methyl orange were studied. Photodegradation process for methyl orange solution at acidic condition showed high removal percentage. 5 mg L-1 methyl orange achieved color removal until 94% for 180 min irradiation in photodegradation process with the presence of C-N-codoped TiO 2 catalyst, and approximately 70% TOC removed as the results of mineralization process. The kinetics reaction rate of photodegradation of methyl orange dye with the addition of C-N-codoped TiO2 followed pseudo-first order represented by Langmuir-Hinshelwood model, the kinetics constant of photodegradation became higher by decreasing the initial concentration of methyl orange. Three organic byproducts of methyl orange formed during photodegradation process was identified by LC-MS/MS system then degradation pathway of methyl orange photodegradation in the presence of C-N-codoped TiO 2 catalyst was proposed.

Costus scaber is an herbaceous species that has hard and cylindrical stems and can reach up to 80 cm in height. It has an inflorescence at the end of the stalk from 6 to 10 cm long, which is surrounded by scaly bracts. Due to this species interesting medicinal potential, the objective of this study was to analyze its cytotoxic and genotoxic effect. The cytogenotoxic and antiprofilerative effect was evaluated using the bioindicator Allium cepa via the direct and indirect method. Fresh Leaves of C. scaber were used for the infusion. Three different concentrations of infusions were elaborated, which were 2, 3 and 4 g/L -1 for each of the different methods, and were analyzed at 24, 48 and 72 hours. Five slides were made per treatment, with 300 cells per slide. Statistical analyses were performed in the R program. The cytotoxic, genotoxic and antiprofilerative effects were found in certain concentrations and at certain exposure periods, and it was also noted that, in general, as the concentration and exposure period increased, the mitotic index decreased and the antiprofilerative effect increased. Several chromosomal abnormalities were found in the infusions analyzed. Therefore, according to the results found, it can be concluded that C. scaber has great potential for genetic improvement of plants in the field of drug production mainly in the field of oncology.
