The morphology of Candida albicans in human candidosis. A light and electron microscopic study

Summary Following the addition of 10 μg/ml of Econazole‐nitrate (2.2 × 10‐5 M), Candida albicans cells were incubated in vitro at 28d̀ C for 30 hours. Numerous lytic areas, usually 50 nm in diameter, occur in the cytoplasm of each cell after 12 hours. An increasing amount of electron dense material, probably containing lipids accumulates in the pathologically changed mitochondria. At that time, aggregation of cytoplasmic components takes place in some cells, resulting in clot‐like formations in ultrathin sections. A 17‐hour exposure to Econazole induces increased lipid accumulation as well as an increase in the number of lysosomes inside bubble‐shaped distorted cells. No more propagating cells are discernible after 30 hours. All cell components have more or less lysed. Comparison with biochemical findings of other authors suggests that, in the primary stage of damage, the imidazole derivatives alter the permeability of the cell envelope. The fungicide penetrating into the cell, causes pathological changes in the cytoplasm and interferes with the RNA and protein synthesis. In the further course of pathogenesis the lipid metabolism is inhibited so that the intracytoplasmic membrane systems are lysed and metabolic end products containing lipids are accumulated in the mitochondria. Zusammenfassung In vitro Zellen von Candida albicans wurden nach Zusatz von 10 μg/ml Econazol‐ Nitrat (2,2 × 10‐5 M) 30 Stunden bei 28°C inkubiert. Nach 12 Stunden erscheinen im Cytoplasma der Zellen zahlreiche, etwa 50 nm große, lytische Bereiche. In den pathologisch veränderten Mitochondrien häufen sich in fortschreitendem Ausmaß elektronendichte, vermutlich lipidhaltige Substanzen an. Zu diesem Zeitpunkt setzt in manchen Zellen eine Aggregation cytoplasmatischer Bestandteile ein, die in Ultradünnschnitten als schollenartige Gebilde erscheinen. 17 Stunden unter Econazol‐Einfluß bewirken in den blasenförmig verformten Zellen außer einer verstärkten Lipidablagerung eine zahlenmäßige Zunahme der Lysosomen. Nach 30 Stunden sind keine vermehrungsfähigen Zellen mehr nachweisbar. Alle Zellbestandteile sind mehr oder weniger stark lysiert. Vergleiche mit biochemischen Ergebnissen anderer Autoren lassen vermuten, daß im Primärstadium der Schädigung durch die Imidazol‐Derivate die Permeabilität der Zellhülle verändert wird. Das in die Zelle eingedrungene Fungizid verursacht pathologische Veränderungen im Cytoplasma und greifi in die RNA‐ und Proteinsynthese ein. Im weiteren Verlauf der Pathogenese wird der Lipidstoff wechsel inhibiert, so daß es zu einer Lyse der intracytoplasmatischen Membransysteme und zu einer Anhäufung lipidhaltiger Stoffwechselprodukte in den Mitochondrien kommt.
