This study evaluated the antibacterial activity of propolis-based toothpastes used as intracanal medication in endodontic treatment. The propolis-based toothpastes were prepared using an extract established in previous studies (identified as A70D and D70D). Calcium hydroxide paste was used as a control. The diffusion-well method on double-layer agar was used in a culture medium of Tryptic Soy Agar. The plates were kept at room temperature for two hours to allow the diffusion of pastes in the culture medium, and then incubated at 35º C for twenty-four hours in aerobiosis and in microaerophilia (S. mutans). After this period, the total diameter of the inhibition halo was measured. The results were analyzed by ANOVA analysis of variance, followed by the Tukey test at p<0.05. The propolis-based toothpastes presented antibacterial activity against 83.3% of the analyzed bacteria. For 66.7% of these bacteria, the propolis-based toothpastes exhibited greater antibacterial activity than calcium hydroxide. The present results allow us to conclude that the experimental pastes A70D and D70D showed good activity against aerobic bacteria, proving more effective than calcium hydroxide.Uniterms: Propolis/antibacterial activity. Endodontics/ treatment.O objetivo deste estudo foi avaliar a atividade antibacteriana de formas farmacêuticas a base de própolis para uso no tratamento endodôntico como medicação intracanal. As formulações de própolis, em forma de pastas, foram preparadas a partir de um extrato pré-estabelecido em estudos anteriores e identificadas como A70D e D70D. Como controle, foi utilizado pasta de hidróxido de cálcio. (26.5 -coagulase negativa) e Staphylococcus epidermidis (6epi). Foi utilizado o método poço difusão em camada dupla de ágar, em meio de cultura Tryptic Soy Agar. As placas foram mantidas à temperatura ambiente por 2 h para permitir a difusão das pastas no meio de cultura, e então incubadas a 35 ºC por 24 h em aerobiose e em microaerofilia (S. mutans). Após este período, foi medido o diâmetro total do halo de inibição. Os resultados foram submetidos ao teste de análise de variância ANOVA seguido do teste de Tukey com p<0,05. As pastas a base de própolis apresentaram atividade antibacteriana contra 83,3% das bactérias analisadas. Para 66,7% das bactérias, as pastas de própolis apresentaram maior atividade

Abstract:We investigated the anti-caries effects of an experimental propolis varnish in vivo, and further tested its toxicity against fibroblasts. Fifty-six SPF female Wistar rats were infected with Streptococcus mutans UA159 (SM) and allocated into four groups (n = 14/group): G1, propolis varnish (15%/PV); G2, chitosan varnish (CV/vehicle); G3, gold standard (GS/Duraphat ® ); and G4, untreated. The animals received a single varnish application on their molars and were submitted to a high cariogenic challenge (Diet-2000, 56% sucrose, and 5% sucrose-added water, ad libitum) for 4 weeks. Total cultivable microbiota and SM were counted, and smooth-surface and sulcal caries were scored. PV, CV and GS cytotoxic effects were tested against fibroblasts. The data were analyzed using ANOVA with the TukeyKramer test (p ≤ 0.05). Total microbiota and SM counts did not differ among the treatments (p = 0.78), or in relation to the untreated group (p = 0.52). PV reduced development of smooth-surface enamel caries compared with the untreated group (p = 0.0018), with no significant difference from GS (p = 0.92); however, the PV effects were no longer observed when the dentin was affected. Neither PV nor GS prevented enamel sulcal lesion onset, but GS significantly reduced the severity of dentinal sulcal lesions (p < 0.0001). No significant difference was observed in fibroblast viability between PV and GS (p < 0.0001).In conclusion, PV prevented smooth-surface enamel caries and showed low cell toxicity. Nevertheless, due to the high cariogenic challenge, its effects were not sustained throughout the experiment. Further studies are encouraged to establish a protocol to sustain the long-term anticaries activity of PV in the oral cavity.

OBJECTIVE: This study investigated the antibacterial action of 30% red propolis, 40% green propolis and 2.5% sodium hypochlorite with irrigation protocols in infected root canals. METHODS: During 60 days, twenty-four root canals were inoculated with E. faecalis. In all experimental groups were performed root canal preparation and two irrigation protocols -passive ultrasonic irrigation and conventional irrigation. In the groups 1, 3, 5 and 7 it were made root canal preparation associated to conventional irrigation with 30% propolis, 40% propolis, 2.5% sodium hypochlorite and distilled water, respectively. In groups 2, 4, 6 and 8 it was made root canal preparation associated with passive ultrasonic irrigation with the solutions described above. The groups 9 and 10 were the controls (negative and positive). Samples of the root canals were collected and immersed in 7 mL of BHI, for 48 hours, at 37ºC. Bacterial growth was analyzed by turbidity of the culture medium. RESULTS: Antibacterial action was observed of 30% red propolis and 2.5% sodium hypochlorite when used passive ultrasonic irrigation and conventional irrigation after 20 minutes, but in 72 hours it was not effective. CONCLUSION: The irrigating agents and protocols were not effective to eliminate the microorganisms of the infected root canals. Clinical Significance: Propolis presents potential for clinical application due to its antimicrobial, anti-inflammatory, antioxidant and low toxicity properties. Key words: Endodontic; Propolis; Sodium hypochlorite Ação antimicrobiana do extrato de própolis vermelho e verde em canais radiculares infectados RESUMO OBJETIVO: Investigar a ação antibacteriana do própolis vermelhos 30%, do própolis verde 40% e do hipoclorito de sódio 2,5% com protocolos de irrigação em canais radiculares infectados. MÉTODOS: Durante 60 dias, vinte e quatro canais radiculares foram inoculados com E. faecalis. Em todos os grupos experimentais foi realizado o preparo do canal radicular e dois protocolos de irrigação -irrigação ultra-sônica passiva e irrigação convencional. Nos grupos 1, 3, 5 e 7 foi feito o preparo do canal radicular associado à irrigação convencional com própolis 30%, própolis 40%, hipoclorito de sódio 2,5% e água destilada, respectivamente. Nos grupos 2, 4, 6 e 8 foi feito o preparo do canal radicular associado à irrigação ultra-sônica passiva com as soluções descritas acima. Os grupos 9 e 10 foram os controles (negativos e positivos). Amostras dos canais radiculares foram coletadas e imersas em 7 mL de BHI, durante 48 horas, a 37ºC. O crescimento bacteriano foi analisado por turvação do meio de cultura. RESULTADOS: Ação antibacteriana foi observada com própolis vermelho 30% e hipoclorito de sódio 2,5% quando utilizado irrigação ultra-sônica passiva e irrigação convencional após 20 minutos, porém, após 72 horas as substâncias não foram efetivas. CONCLUSÃO: Os agentes de irrigação e os protocolos não foram efetivos para eliminar os microorganismos dos canais radiculares infectados.
