Seven arylideneacetophenones: 1-(4-chlorophenyl)-3-phenyl-2-propen-1-one (1a), 1-(4-nitrophenyl)-3-phenyl-2-propen-1-one (1b) , 1-(4-methoxyphenyl)-3-phenyl-2- propen-1-one (1c) , 1-(4-methylphenyl)-3-phenyl-2-propen-1-one (1d) , 1-(4- chlorophenyl)-3-(4-methoxyphenyl)-2-propen-1-one (1e) , 1-(4-methylphenyl)-3-(4- methoxyphenyl)-2-propen-1-one (1f) and 1,3-di(4-methoxyphenyl)-2-propen-1-one (1g) were synthesized from the corresponding substituted acetophenones and benzaldehydes in presence of sodium hydroxide in aqueous ethanol. Triple bond analogues of polyunsaturated fatty acids irreversibly inactivate lipoxygenases. During the inactivation the inhibitors are converted enzymatically [Kuhn, H., et al. (1984) Eur. J. Biochem. 139, 577-5831. Since the converted inhibitor molecules may hold important information about the inactivation mechanism, we have determined the structure of the product that is formed during the irreversible inactivation of soybean lipoxygenase-1 by octadeca-9,12-diynoic acid (ODYA), the triple bond analogue of linoleic acid. This product is formed only in the presence of Fe(II1)-lipoxygenase-1 and 0 2 . It was purified by C18 solid phase extraction and reversed phase HPLC and was identified with UV, IR, and NMR spectroscopic and mass spectrometric techniques as the novel lipoxygenase product,

Synthesen ungesättigter Pyrazol‐Derivate, II¹⁾ Reaktionen von 1.5‐Diphenyl‐pentadiin‐on‐(3) und 1.5‐Diphenylpentenin‐on‐(3) mit Hydrazin bzw. Hydroxylamin

Unter schwach sauren Bedingungen addiert sich Hydrazin an die Carbonylgruppe von 1.5-Diphenyl-pentadiin-(1.4)-on-(3) (1) zum Ketazin 2, unter neutralen und basischen Bedingungen an die Dreifachbindung unter Bildung des Athinyl-pyrazol-Derivates 3. Uberschussiges Hydrazin addiert sich an dessen Dreifachbindung, wobei sich die isomeren Hydrazone 6 und 7 im Verhaltnis 2 : 3 bilden. Aus Hydroxylamin und 1 entsteht primar das Hydroxyisoxazolin-Derivat 10, welches beim Erhitzen in das Isoxazol 12 ubergeht. 1.5-Diphenyl-pentenin-on-(3) (13) liefert mit Hydrazin das Gemisch der isomeren Hydrazin-Derivate 14 des Phenathylpyrazols, mit Hydroxylamin nur eines der beiden moglichen Hydroxylamin-Derivate (15) des Phenathyl-isoxazols. Die Ergebnisse erlauben es nicht, den Ort der Primaraddition der Base festzustellen.Wir 1) haben kurzlich drei Synthesewege zu ungesattigten Pyrazolen aufgezeigt und berichten nun uber die Reaktion von Hydrazin (bzw. Hydroxylamin) rnit substituiertem Diathinylketon (Weg B in Lit.1)) bzw. rnit einern En-inketon, um u. a. die elektrophile Reaktivitlt aktivierter Doppel-und Dreifachbindungen zu vergleichen.Bekanntlich 2) addieren Athinylketone Hydrazin unter Bildung von Pyrazol-Derivaten.Methylhydrazin reagiert mit 1 -Phenyl-butin-( l)-on-(3) zum Gemisch von 1.5-Dimethyl-3phenyl-und 1.3-Dimethyl-5-phenyl-pyrazol3), wahrend aus Phenylpropiolaldehyd und Phenylhydrazin ein Phenylhydrazon entsteht4), welches sich therrnisch nicht in das entsprechende Pyrazol-Derivat iiberfuhren IaBt5).
