The effects of desferrioxamine on cisplatininduced lipid peroxidation and the activities of antioxidant enzymes in rat kidneys

Abstract: Cisplatin-induced nephrotoxicity is associated with an increase in lipid peroxidation and oxygen free radicals in rat kidneys. In this study, the effects of desferrioxamine were compared to vitamin C and E on cisplatin-induced lipid peroxidation and antioxidant enzyme activities in rat kidneys. Rats were divided into five groups, with 15 Wistar rats in each group. In the control group, rats received 1 mL/100 g isotonic saline solution intraperitoneally (i.p.). In Group II, 10 mg/kg cisplatin i.p. was … Show more

“…Results from the current work further support reports from several studies that provided evidence that the cellular events in cisplatin-mediated nephrotoxicity are a consequence of reactive oxygen species (ROS) generation, which produces oxidative renal damage [Yilmaz et al, 2004;Kadikoylu et al, 2004]. Cisplatin in the present study produced significant reduction of the activities of plasma catalase (an antioxidant enzyme), plasma total antioxidant capacity accompanied by increased lipid peroxidation compared with the control rats.…”

“…In fact, there are evidences of the renal increase in NO production in cisplatin nephrotoxicity (27). The O 2 •-increase in cisplatin-nephrotoxicity may be simply consequence of the mitochondrial dysfunction (4) and the decrease in superoxide dismutase activity (28). One of the most important intracellular antioxidant systems is the glutathione redox cycle.…”

“…Amifostine decreases generation of free oxygen radicals and might have a protective effect on adriamycin toxicity (Singal and Tong 1988;Dorr 1996;Kang et al 1996;Samelis et al 1998;Kocak et al 2003;Kadikoylu et al 2004). Adriamycin-induced hematological and testicular toxicities, cataract formation, and cisplatininduced neuro-, nephro-and ototoxicities may be either prevented or decreased by administration of amifostine before cytotoxic chemotherapy in experimental and clinical studies (Spencer et al 1995;Santini and Giles 1999).…”
