The effects of calcipotriol and methylprednisolone aseponate on bcl‐2, p53 and ki‐67 expression in psoriasis

Abstract: The results of this study provide evidence that both calcipotriol and MPA decrease the p53 and ki-67 expression and increase bcl-2 expression. However, it should further be elucidated if these changes were the common behaviour of psoriatic keratinocytes to any antipsoriatic medication.

“…Simultaneously, the complex preparation, containing calcipotriol and betamethasone, contributed to a decrease in Ki-67 antigen expression by 5% at week 4 and by 74% at week 6 of the treatment [29]. According to Adişen et al, after six-week therapy with calcipotriol, the Ki-67 antigen expression, which was high before, significantly decreased [30]. Similar findings were observed with regards to methylprednisolone aceponate.…”

“…4 Ki-67 is an established marker of cell proliferation. 6 It is highly expressed in psoriasis and correlated with the clinical severity of disease. 6 It is highly expressed in psoriasis and correlated with the clinical severity of disease.…”

“…For cell proliferation, Ki‐67 protein is required, and it is used as a proliferation indicator. Cells expressing Ki‐67 in normal skin are 1–3% and are mostly found in the basal and suprabasal layers . The high level of Ki‐67 in oral LP confirms that it may possibly have a secondary proliferative disorder due to the cyclic fragmentation of cells leading to proliferation .…”

“…Keratinocytes in the superficial epidermis regulate or stabilize the proliferation of basal cells. Furthermore, physiological apoptosis has been demonstrated in the germinative region of the nonpathogenic epidermis . Apoptotic keratinocytes can be seen in diseases such as psoriasis and LP .…”
