Research was conducted at Al-Qasim Green University in the Plant Tissue Culture Laboratory in the Department of Horticulture and Landscaping in the College of Agriculture from November 2021 to May 2022 with the aim of comparing two types of cytokinins: BAP at a concentration of 2.0 mg/L-1 and TDZ at a concentration of 0.2 mg/L-1 and two concentrations Iron (2.5-5 mg/L-1). To know their effect on the average of shoot multiplication in vitro of the root and its content of protein and macro (N.K) and Micro (iron) elements. The research was implemented as a factorial experiment using a complete random design (CRD) with two factors: cytokinin and nano iron. The results indicated a superiority of 0.2 mg.L-1 TDZ in each of the Multiplication shoots (4.667 shoots/explant ) and the content of shoots of iron (54.66 mg.kg-1), while BAP at a concentration of 2.0 mg/L-1 was excelled on the content of shoots of nitrogen, potassium and protein (1.778%, 1.947%.) respectively. As for nano iron at a concentration of 2.5 mg. L-1 was significantly excelled in the average of multiplication shoots (5 shoots/explant ), nitrogen (1.765%) and protein (10.98%). Specifically, the 5 mg dose. Potassium and iron (1.692% and 55.0 mg.kg-1) were most abundant in L-1.

Date palm (Phoenix dactylifera L.) is one of the most important economic plants which belongs to the Arecaceae family in arid and semi-arid areas. The genetic map of the date palm genome is incomplete, which hindered the expansion of molecular studies. Bioinformatics is a way that the researcher compares different plant species to find similarities and differences between them. MYB is a transcription factor family that has well-known functions in many plants, especially Arabidopsis. DNA-binding domains are pieces of peptides (a group of amino acids) that are folded independently and are highly attracted to DNA. The study aims to identify the DNA-binding domains for the MYB family in Date Palm to diagnose drought-related genes in the date palm genome through bioinformatics. The main approach followed in the research is to use a set of approved sites to align members of MYB proteins in Arabidopsis with the date palm genome to determine the most closely related MYB proteins in date palms. The results indicated a group of date palm proteins, which considered candidate proteins or genes to have an effective role in resisting or reducing the negative effects of water stress on date palms. Also, numbers of palm proteins have the same alignment effect and DNA-binding domains, which means that they may be close to each other in function and structure.

Background Mass propagation of date palm has attracted the interest of commercial producers. However, this technique still faces many obstacles that hinder production. This study investigated the effect of chitosan (CHT) at various concentrations for the possibility to apply it in combination with thidiazuron (TDZ) on the growth and development of tissue cultures of Barhee cultivar. Results The results showed that CHT and TDZ on in vitro proliferation of Barhee date palm cultivar were significant. The highest response rate and the number of shoots per jar were found in MS media supplemented with 15 mgL–1 CHT and 0.5 mgL–1 TDZ combination. Furthermore, we found that the combined application between 20 mg L−1 CHT+ 1.0 mg L−1 TDZ resulted in the highest shoots content of endogenous IAA, compared with other treatments. At the same time, the data revealed that the maximum cytokinins (CKs) content of shoots occurred in a medium supplemented with 15 mg L−1 CHT and 0.5 mg L−1 TDZ. The genetic stability of the discussed micropropagation protocol was confirmed in this study by DNA-based technique RAPD (random amplified polymorphic DNA). The results may indicate that the micropropagation protocol developed in this research paper was appropriate and applicable for producing genetically stable date palm cv Barhee plants. Conclusion Applying the strategy of culture treatment with (CHT) and (TDZ) can be valuable for improving the propagation of date palm cv Barhee in vitro.
