The effect of explant material on somatic embryogenesis of Cyclamen persicum Mill

Abstract: Somatic embryos of Cyclamen persicum Mill. could be produced through a callus phase from juvenile explant material including anthers, ovaries and zygotic embryos. The auxin 2,4-D (1.0-1.5 mg 1-1) and coconut milk (10% v/v) in MS medium were important factors for the induction of somatic embryogenesis. Somatic embryos germinated into plantlets in MS medium without growth regulators. The plants grew well in the greenhouse and flowered normally. The plants were phenotypically identical to the mother plants with a… Show more

“…Besides reports on organogenesis (reviewed by Takamura & Miyajima, 1997;Dillen et al, 1996), efficient protocols for somatic embryogenesis have been published (Wicart et al, 1984;Fukui et al, 1988;Otani & Shimada, 1991;Kiviharju et al, 1992;Kreuger et al, 1995;Takamura et al, 1995;. While some protocols included seedling tissues as starting material (Kreuger et al, 1995;Takamura et al, 1995), we have concentrated on the method starting from somatic tissue in ovules .…”

“…In cyclamen (Cyclamen persicum) somatic embryogenesis is an effective vegetative in vitro propagation method (Kiviharju et al 1992;Kreuger et al 1995;Schwenkel and Winkelmann 1998;Winkelmann et al 2000). Mass propagation using embryogenic suspension cultures was found to be even more effective if bioreactor culture was applied (Hohe et al 1999a(Hohe et al ,b, 2001.…”

“…Since cyclamen cannot be propagated from cuttings, in vitro culture techniques have to be applied. Many genotypes can be multiplied efficiently via somatic embryogenesis (Wicart et al 1984;Otani and Shimada 1991;Kiviharju et al 1992;Kreuger et al 1995;Takamura et al 1995;Schwenkel and Winkelmann 1998;Winkelmann and Serek 2005).…”
