Changes in Cu concentration in the livers of adult Scottish Blackface (B), Welsh Mountain (W) and B x W ewes fed diets containing 4, 9, 17 and 29 mg Cu/kg D.M. were measured over a 28-week period. Measurement continued during two further periods of 18 weeks when first all sheep were fed the diet containing 4 mg Cu/kg D.M. and finally, when 4 mg Mo/kg D.M. was added to this diet.During the first period the increase on all diets was least in B ewes and, apart from the 4 mg Cu/kg D.M. diet, greatest in W ewes; this difference increased as the Cu in the diet increased. The rate of increase for all ewes diminished towards the end of the period. All breed types retained proportionately less Cu in their livers as the Cu in the diet increased; at one extreme B ewes fed 17 or 29 mg Cu/kg D.M. retained equal amounts.When during the second period all sheep were fed the 4 mg Cu/kg D.M. diet the Cu concentration in their livers decreased, the rate of decrease being greatest for ewes whose mean concentration in the liver was highest, but at similar concentrations the decrease was greater in B than W ewes. During the final period the decrease was faster and was again associated with the mean Cu concentration, but the decrease was not significantly affected by breed.Throughout the trial, W ewes had the highest concentration of Cu in their plasma, whilst B ewes and ewes fed the 4 mg Cu/kg D.M. diet throughout had the lowest concentrations both in plasma and in their kidneys at slaughter.The results show that breeds differ in their ability to retain Cu from the diet in their livers. The magnitude of these differences depends on the intake of Cu and Mo from the diet and is affected by the concentration of Cu in the liver. Breed differences in the absorption and endogenous loss of Cu are suggested.

RESUMO.-Devido à carência de informações sobre o tema e à ausência de estudos que confrontem as técni-cas de biópsia hepática em ovinos, foi desenvolvido um estudo comparativo entre oito técnicas de biópsia nesta espécie. Neste estudo foram utilizadas oito borregas (17,95kg ± 2,71) mestiças (Santa Inês) em procedimentos seriados por oito semanas consecutivas e avaliados os aspectos clínicos, os achados de patologia clínica, o peso das amostras hepáticas, a qualidade histológica, o número de veias centrolobulares e espaços-porta e a presença de artefatos no corte histológico de amostras hepáticas obtidas através de seis técnicas percutâneas e duas técnicas videolaparoscópicas. Para as técnicas percutâneas empregaram-se dois tipos de agulhas, Men- Due to lack of information about the subject and lack of studies that confront the techniques of liver biopsy in sheep, a comparative study between eight biopsy techniques in this species was developed. In this study, eight crossbred (Santa Inês) ewe lambs (17.95 kg ± 2.71) were used in serial procedures for eight consecutive weeks. Liver samples obtained through six percutaneous techniques and two videolaparoscopic techniques were analyzed. The clinical aspects, clinical pathological findings, hepatic sample weights, histology quality, number of centrilobular veins and portal spaces, and the presence of artifacts in the histology slices were recorded. For the percutaneous techniques, two types of needles were used, modified Menghini (M) and semi-automatic tru-cut (T). They were used in combinations with blind biopsies, guided by ultrasound (US) and monitored by video laparoscopy (VL) biopsies. In the videolaparoscopy procedures, Blakesley biopsy forceps and the method of liver fragment resection by VL were used. No relevant hematological or biochemical alterations were found and the clinical manifestations were slight and transitory. In a general way, the use of US and VL in percutaneous biopsies did not yield a significant improvement in weight and histology quality of the liver samples; however, the videolaparoscopic techniques allowed a wider access to the liver than the percutaneous techniques. The quality of the samples obtained through both VL techniques were equivalent; however, the resection technique enabled the collection of heavier samples, but determined the formation of perihepatic adherences. More difficulty in fragment retrieval, more fluctuation in weight and less quality were found in samples obtained with M needles when compared to T needles.INDEX TERMS: Liver biopsy, videolaparoscopy, ultrasonography, sheep.

Experiments were conducted to test the commonly held assumption that analysis of a portion of rat liver is representative of the elemental concentration of the whole organ. Male Sprague-Dawley rats (initial body weight approximately 250 g) fed a chow diet or weanling male Long-Evans rats (initial body weight approximately 50 g) fed a semipurified diet with or without copper in the mineral premix were sacrificed after 4 wk on their respective diets and livers were dissected into seven portions representing major anatomical divisions of this organ. Elemental analyses by atomic absorption spectroscopy (calcium, magnesium, iron, zinc, copper, manganese), atomic emission spectroscopy (sodium, potassium), or colorimetric assay (phosphorus) demonstrated no statistically significant differences in composition of these nine elements among anatomical regions of liver. Dietary copper deficiency led to equivalently reduced copper concentration in all portions of rat liver and did not cause any other significant alterations in liver composition of these nine elements within the 4 wk of these studies. These results confirm the validity of the common assumption that analysis of a portion of rat liver can be representative of the elemental composition of the whole organ. This conclusion will allow more analyses to be performed on fewer animals, thereby reducing animal use and reagent costs without sacrificing analytical accuracy.
