“…Dinitrochlo robenzene (DNCB) penetrates the epidermis very rapidly: specific fluorescence was observed throughout the epider mis 1 min after epicutaneous painting with DNCB in the guinea pig [1]. Most of the DNCB is absorbed through the skin via the venous and lymphatic systems.…”

“…Human studies reveal broadly similar findings, with DNCB localized in the upper epidermis at 1 h after chal lenge and detectable on sweat duct and hair follicle epithelia at 3 h [3], Studies using radioactive nickel applied to the skin surface showed 65% absorption within 24 h. there being no difference between nickel-sensitive and non-nickel-sensitive subjects [4], DNCB has been demonstrated on epidermal cell membranes at 3 h in epidermal sheets from guinea pigs [5], Cytoplasmic localization has also been demonstrated in epidermal cells [1], including both keratinocytes and Langerhans cells [6. 7], DNCB-reactive cells are noted in the upper dermis as early as 6 min in animal studies [1] and at 6 h in humans [3].…”

“…Our previous investigation [7], in which localization of 2,4-dinitrophenyl (DNP) groups in the skin of guinea pigs following painting with 2,4-dinitrochlorobenzene (DNCB) was examined by the immunofluo rescent method using anti-DNP antibody, showed that DNP groups were distributed on/in epidermal cells. The cells in which DNP groups were detectable (DNP cells) were shown to account for approximately 90% of the epidermal cells [9], This indicates that the majority of DNP cells are kératino cytes.…”

“…This reaction results in the formation of dinitrophenylated proteins which are recognized as foreign by the immune system (20). It has been shown previously that DNCB readily penetrates the skin of humans and monkeys (3)and that followingapplication ofDNCB, dinitrophenylated proteins can be detected in draining lymph nodes (19). Because DNCB reacts with structural as well as soluble proteins, it acts as a persistent antigen which favors formation of cellular as opposed to humoral immunity.…”
