Distribution of 2,4-Dinitrophenyl Groups on the Epidermal Langerhans Cells of Guinea Pigs following Skin Painting with 2,4-Dinitrochlorobenzene

Abstract: It has been postulated that epidermal Langerhans cells (LC) are essential for induction and elicitation of contact sensitivity (CS) to simple chemical allergens. Although the precise role of LC in CS has not been determined, recent reports have focused on a function for LC in the presentation of antigen to T lymphocytes. To investigate this possibility, the distribution of allergen on the epidermal LC of inbred strain 13 guinea pigs was observed by the immunofluorescent method using antibodies against la antig… Show more

“…It has been shown by our present and previous works (3,4) that DNCB applied to animal skin enters the epidermis where it binds cell membrane components, not only of keratinocytes and Langerhans cells, but also of Thy-l + ECs. One of the important questions raised by our studies is what role these haptenated ECs play in the development of CS.…”

“…Our previous investigations (3,4), in which localization of 2,4-dinitrophenyl (DNP) groups in the skin of guinea pigs following painting the skin with 2,4--dinitrochlorobenzene (DNCB) was examined by the immunofluorescent (IF) method using antibody against DNP groups in order to clarify the mechanism of contact sensitivity (CS), showed that DNP groups were distributed on/in keratinocytes and Langerhans cells. The object of the experiments reported here was to determine whether DNP groups are also localized on Thy-I+ cells in the epidermis of mice following skin painting with DNCB.…”

“…Thus presentation by acces sory cells to T lymphocytes might occur without intracellu lar processing of the antigen. Oka et al [6] also observed an equal affinity of keratinocytes and Langerhans cells for epicutaneously applied DNCB.…”

“…The epicutaneous application of DNCB is a well-established model for the study of cell-mediated immunity in both humans and laboratory animals. 4,8,22,25,26,32 Contact hypersensitivity may be considered to have two stages. There is an initial stage of sensitization resulting in the production of memory T cells and then, upon reexposure to the allergen, an elicitation phase characterized by the infiltration of the epidermis and dermis at the site of contact with mononuclear cells.…”
