The Determination Of Morphine in Opium and Some of Its Galenical Preparations

Abstract: A method of assay for morphine in opium depending upon precipitation of the dinitrophenylether with 1‐fluoro‐2:4‐dinitrobenzene has been critically examined. The morphine is first separated from other alkaloids and extraneous matter by elution from an alumina column with an organic solvent and subsequent extraction with alkali. The method, which is rapid, has been applied to many galenical preparations and, in cases where the pharmacopoeial method based on the colorimetric nitroso reactions may give high resul… Show more

“…Besides solving well-known problems associated with the classical solvent extractionsolvent removal method, the S P E technique is particularly attractive as it lends itself to coupling with chromatographic systems for on-line applications (14). Another impressive feature of the SPE technique is that small sorbent traps can be deployed in the field by using newly available submersible instrumentation (15,16). In this way, combined sampling, extraction, and preconcentration are done at the sampling site, thus eliminating most contamination and handling problems connected to the sample collection.…”

“…Besides its use in structural analysis, FDNB has been employed for the spectrophotometric determination of amino acids and primary and secondary amines (3-9), amino acid nitrogen in plasma and 0003-2700/89/0361-1358$01.50/0 urine (10, l l ) , isoniazid (12), various aminoglycoside antibiotics (13), phenols (14), and the enzyme amidase (15). It has also been used in the gravimetric determination of morphine (a phenolic alkaloid) (16) and other phenols (In, as a derivative reagent in gas-liquid chromatography (GLC) for phenols (18) and amines (19), in high-performance liquid chromatography (HPLC) for amines and aminoglycosides (20)(21)(22), and in thin-layer chromatography (TLC) and mass spectrometry for amines (23).…”
