The association analysis of lncRNAHOTAIRgenetic variants and gastric cancer risk in a Chinese population

Abstract: The HOX transcript antisense intergenic RNA (HOTAIR), a well-known long noncoding RNA, is involved in pathogenesis and progress of multiple tumors. Its ectopic expression and biological functions have been observed in gastric cancer. In this study, we conducted a two-stage case-control study to evaluate whether genetic variations of HOTAIR were associated with gastric cancer risk. We identified that a single nucleotide polymorphism (SNP) rs4759314 was significantly associated with the increased gastric cancer … Show more

“…In current study, we detected significant associations between the rs4759314 and rs7958904 and EOC susceptibility among Chinese population, some studies have also demonstrated significant associations between rs7958904 and decreased risk of colorectal cancer [7], rs4759314 with the increased gastric cancer risk [20]. The major strength of this study is the use of a large sample size to study genetic predispositions, which are less likely to be affected by confounding factors.…”

“…The lncRNA HOTAIR could modify the progress of carcinogenesis [7, 12, 20]. HOTAIR (for HOX transcript antisense RNA), was located on chromosome 12: 53.96–53.97 Mb [15].…”

“…A possible mechanism underlying the association of these alleles to increased susceptibility is through the regulation of the expression of HOTAIR itself. Polymorphism rs4759314 resides in an intronic promoter region, which was found to influence the activity of this promoter and expression of HOXC11 gene [23]. For polymorphism rs4759314 , increased HOTAIR expression originated from the G (relative to the A) allele in luciferase reporter assays.…”

“…SNP rs4759314 was also significantly associated with increased risk of GC. Further functional experiments revealed allele-specific effects on HOTAIR expression in GC tissues; individuals with the AG genotype showed higher HOTAIR expression compared with those carrying the AA genotype, Nevertheless, whether HOTAIR SNP rs4759314 could act as a biomarker for predicting GC occurrence has yet to be determined [83]. Overall, accumulating evidence indicated that HOTAIR, which is present in tissues, blood, and gastric juice, was dysregulated in GC and related to tumorigenesis, metastasis, and progression, exhibiting potential as a biomarker for the diagnosis and prognosis of GC.…”
