Association of HOTAIR polymorphisms rs4759314 and rs920778 with cancer susceptibility on the basis of ethnicity and cancer type

Abstract: Polymorphisms in the HOX transcript antisense intergenic RNA (HOTAIR) have been recently associated with susceptibility to different cancers. Here, a meta-analysis was performed to derive a more precise estimation of the involvement of HOTAIR polymorphisms in cancer development. Data from cases (n = 7,772) and controls (n = 9,075) were extracted from eligible studies (n = 10) identified in a comprehensive literature search conducted in PubMed, Embase, and the Web of Science databases through January 20, 2016. … Show more

“…However, the results remain contradictory and inconclusive, especially in different geographical location and ethnicity groups. In 2016, three similar meta-analyses involving eight articles explored the association of HOTAIR polymorphisms with several kinds of cancers among worldwide population [ 34 36 ]. However, none of them drew a conclusion for Chinese population.…”

“…Genetic polymorphism rs4759314 from the AA genotype to the GG genotype is significantly associated with susceptibility to gastric cancer [19]. The TT genotype at the rs1899663 site increases the risk of breast cancer compared with the GG homozygotes in the Chinese population [20]. Therefore, these four sites are closely associated with the onset, progression, and poor prognosis of various cancers [21][22][23][24].…”

“…These observations are likely to reflect tumor-specific carcinogenic processes that are not shared among these tumor types, and also due to ethnic differences among studies. In fact, a recent meta-analysis showed that HOTAIR SNP rs920778 was associated with gastric cancer and esophageal squamous cell carcinoma, but not with breast cancer, in Asians but not in Caucasians [38]. Nonetheless, limitations in the sample size of our and others' studies must not be ruled out, warranting further studies in larger cohorts to validate these findings.…”
