In nature, orchid seed germination is obligatory following infection by mycorrhizal fungi, which supplies the developing embryo with water, carbohydrates, vitamins, and minerals, causing the seeds to germinate relatively slowly and at a low germination rate. The nonsymbiotic germination of orchid seeds found in 1922 is applicable to in vitro propagation. The success of seed germination in vitro is influenced by supplementation with organic compounds. Here, we review the scientific literature in terms of the contents and role of organic supplements in promoting seed germination, protocorm development, and seedling growth in orchids. We systematically collected information from scientific literature databases including Scopus, Google Scholar, and ProQuest, as well as published books and conference proceedings. Various organic compounds, i.e., coconut water (CW), peptone (P), banana homogenate (BH), potato homogenate (PH), chitosan (CHT), tomato juice (TJ), and yeast extract (YE), can promote seed germination and growth and development of various orchids. They also stimulate seedling development, formation of protocorm-like bodies (PLBs), plantlet growth, and multiple shoot formation. The addition of organic compounds to culture media, individually or in combination, accelerates seed germination and seedling development. Different types and concentrations of organic nutrients are needed for the success of in vitro cultures, depending on the species and genotype.

Phalaenopsis merupakan anggrek yang mempunyai nilai ekonomis tinggi karena warna yang menarik. Permintaan selalu meningkat sehingga tehnologi kultur jaringan sangat dibutuhkan dalam perbanyakan anggrek ini. Perbanyakan tanaman anggrek menggunakan sumber eksplan protocorm-like body (PLB) merupakan salah satu cara yang tepat untuk menghasilkan anggrek dalam jumlah banyak. Penelitian ini bertujuan untuk mendapatkan kombinasi NAA dan Kinetin terbaik pada perkembangan PLB di kultur cair serta PLB yang disubkultur di media regenerasi. Bahan tanam menggunakan PLB umur 2 bulan yang ditanam pada media Vacin & Went (VW) cair dengan perlakuan kombinasi NAA dan Kinetin masing-masing pada konsentrasi 2,5 mg/L, 5 mg/L dan 7,5 mg/L. Media regenerasi menggunakan VW padat dengan suplemen 15% air kelapa, 5% pisang, 5% kentang dan 0,2% arang aktif. Percobaan menggunakan Rancangan Acak Lengkap dengan tiga kali ulangan. Hasil penelitian menunjukkan bahwa kombinasi NAA dan kinetin tidak berpengaruh terhadap jumlah dan berat PLB. Sementara itu, warna PLB hampir semua menunjukkan warna yang sama yaitu hijau kekuningan (5GY 7/6) hanya pada kombinasi perlakuan NAA 5 mg/L + Kinetin 7,5 mg/L terjadi klorosis berwarna kuning pucat (5Y8/4). Perubahan warna planlet terjadi dari hijau kekuningan terang (7.5GY8/8) menjadi hijau kekuningan (7.5GY7/6) dan hijau kekuningan tua (7.5GY6/10) masing-masing pada awal aklimatisasi, serta pada dua dan empat minggu. Bibit anggrek tumbuh dengan baik dan menjadi tanaman dewasa setelah dipindahkan pada media moss hitam.

Effective and rapid in vitro plantlet regeneration from protocorms of Cymbidium aloifolium using chitosan, coconut water and potato extract were investigated. Two experiments were performed using twelve week old protocorms of size 2-3 mm. In the first experiment, the protocorms were propagated on solid New Dogashima (ND) medium supplemented with various concentrations of chitosan (0, 0.05, 0.10 and 1.00 mg/l). For the second experiment, the protocorms were cultured on the same medium supplemented with coconut water (0, 100, 150 and 200 ml/l) and potato extract (0, 5, 10 and 15 g/l). Survival and response percentages, shoot number and length, root number and length, leaf number and fresh and dry weight were collected after 10 weeks of culture. The results showed that 0.05 mg/l chitosan treatment was the most effective for protocorm growth, producing the highest shoot number and length, root number and length, leaf number, and fresh and dry weight. The combination of coconut water and potato extract treatments revealed that 150 ml/l coconut water alone was the best for plantlet regeneration and growth, shoot length, root length, and fresh and dry weight. This treatment gave significantly different results from the other treatments. Findings will provide useful information for other in vitro commercial or conservation orchid propagation programs.
