Temporal expression of growth factor genes during myogenesis of satellite cells derived from the biceps femoris and pectoralis major muscles of the chicken

Abstract: The expression of mRNAs for transforming growth factors (TGF-beta2, myostatin, activin-B, and follistatin), insulin-like growth factors (IGF-I and -II), and fibroblast growth factor (basic, bFGF) was investigated in satellite cells derived from chicken pectoralis major (PM) and biceps femoris (BF) muscles in the stages from initiation of proliferation to fusion. These growth factor gene cDNAs were synthesized by reverse transcriptase polymerase chain reaction (RT-PCR). No myostatin, activin-B, follistatin or b… Show more

“…before myoblast fusion and the appearance of myotubes. Several studies have described a similar expressional pattern respectively in chicken satellite cells and C2C12 myoblasts [11,24,43]. However, as in our satellite cell cultures, myostatin up-regulation was observed in C2C12 when fusion was already established [24].…”

“…In humans and in chickens, there is no difference between fast and slow muscles in terms of their myostatin expression in vivo and in vitro [43,46]. In rodents, myostatin mRNA and protein levels are higher in fast than in slow muscles [20,22].…”

“…In contrast, IGF-II gene expression was decreased in turkey satellite cells from proliferation to differentiation (Ernst et al, 1996). In addition, IGF-I expression was not detected in humans (Crown et al, 2000) or avian (Kocamis et al, 2001) satellite cells. The expression patterns of IGF for in vitro muscle cells suggested autocrine regulation of myogenesis by IGFs (Wang et al, 2011).…”

“…For instance, transforming growth factor-b (TGFb) family members control diverse processes in the myoblast and satellite cells ranging from proliferation and fusion to differentiation, and the patterns of expression change dynamically in regenerating muscles to control sequential stages of muscle repair (Kollias and McDermott 2008). Illustrating a switch from an adaptive to a maladaptive mechanism, functional levels of the TGFb family member myostatin, a negative regulator of muscle mass, decline during active regeneration to permit satellite cell activation (Kirk et al 2000;Kocamis et al 2001). However, in chronic myopathic states, myostatin as well as TGFb itself are elevated coincident with fibrosis, and recent interventions to antagonize signaling have been noted to restore muscle function in mouse models of muscular dystrophies (Minetti et al 2006;Cohn et al 2007).…”
