Targeting lung cancer stem‐like cells with TRAIL gene armed oncolytic adenovirus

Abstract: Lung cancer stem cell (LCSC) is critical in cancer initiation, progression, drug resistance and relapse. Disadvantages showed in conventional lung cancer therapy probably because of its existence. In this study, lung cancer cell line A549 cells propagated as spheroid bodies (named as A549 sphere cells) in growth factors-defined serum-free medium. A549 sphere cells displayed CSC properties, including chemo-resistance, increased proportion of G0/G1 cells, slower proliferation rate, ability of differentiation and… Show more

“…ABCG2 gene silencing using short hairpin RNA (shRNA) is a promising therapeutic approach for overcoming drug resistance [12][13][14] . Nanoparticle-mediated gene delivery has demonstrated enhanced transfection efficacy due to the structure, size, and surface properties of the nanoparticles 15,16 , allowing them to readily accumulate in and be retained by solid tumors through the enhanced permeability and retention (EPR) effect 17,18 .…”

“…We also discovered that the antiproliferation effects of Dandelion extracts relates to expression of TRAIL and increased of ROS in cells. Indeed, the potential of using TRAIL to destroy putative cancer stem cells has been recognized by others (Chandrasekaran et al, 2014;Yang et al, 2015). Increasing TRAIL expression has previously been shown to promote cancer cell death and to decrease tumor and metastasis development in vivo.…”

“…Targeting IGFBP-4 may lead to the inhibition of lung cancer stem cell growth. As lung cancer stem cells are associated with chemoresistance, progression and metastasis of lung cancer (25)(26)(27), this may lead to the long-term inhibition of lung cancer development. This may be investigated in a future study.…”
