Complexes of Co(II), Ni(II), Cu(II) and Zn(II) with N-picolinoyl-N H -thiobenzoyl hydrazine (H 2 L) have been prepared and characterized. Based on elemental analyses, infrared, magnetic, electronic, ESR and FAB mass spectra, squareplanar structures for [Cu(L)] and [Ni(HL)Cl], an octahedral structure for [Co(L)(H 2 O) 2 ] and a mixed octahedral7square-planar structure for [Ni(L)(H 2 O)] have been proposed. H 2 L and its soluble complexes have been screened against several bacteria and fungi.

Dibenzobarallene (1) was utilized as the key intermediate for the synthesis of some new 2-substituted 1,4-dioxo-3,4,4a,5,10,10a-hexahydro-1H--5,10-[1',2']-benzenobenzo[g]phthalazine: 2, 5a-d, 8a-c and 10. Condensation of 2 with benzaldehyde or anisaldehyde gave the corresponding acrylonitrile derivatives 3a and b, respectively. Thiophene derivatives 4a and b were obtained via the Gewald reaction of 2 with cyclohexanone or cyclopentanone, respectively. Treatment of 5d with acetyl chloride or p-toluenesulfonyl chloride afforded the corresponding esters 6 and 7, respectively. Cyclization of 8a-c with formalin afforded the corresponding triazine derivatives 9a-c. Ring opening of 10 with sodium hydroxide gave the corresponding triazole derivative 11, which when alkylated with pentyl bromide afforded the pentylthio derivative 12. Representative compounds of the synthesized products were established and evaluated as antibacterial agents.

We describe the synthesis and evaluation of N-acylhydrazone compounds bearing different electrondonating groups in one of its aromatic rings, obtained using a four-step synthetic route. IC 50 values against pathogenic fungi and bacteria were determined by serial microdilution. Compounds showed low activity against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. By contrast, a derivative with a meta-oriented electron-donating group showed significant activity (IC50) against Candida albicans (17 mM), C. krusei (34 mM) and C. tropicalis (17 mM). Results suggest this is a promising leadcompound for synthesis of potent antifungal agents.Keywords: N-acylhydrazones; antibacterial activity; antifungal activity. INTRODUÇÃOA pesquisa por novos antimicrobianos constitui uma das estratégias para evitar o insucesso nos tratamentos anti-infecciosos, causado pela emergência de microrganismos resistentes. Embora o uso inadequado desses fármacos seja um dos principais fatores para o surgimento de linhagens resistentes, sabe-se que a busca por fármacos inovadores que atuem em rotas bioquímicas ainda não exploradas representa um caminho importante para o sucesso da quimioterapia antibacteriana e antifúngica. Com base no exposto, e tendo em vista o potencial antimicrobiano de substâncias que contenham a subunidade hidrazona, nosso grupo de pesquisa sintetizou uma série de cinco acilidrazonas e avaliou, pela primeira vez, o perfil de sensibilidade de bactérias e fungos patogênicos de linhagens padronizadas frente a esses derivados. RESULTADOS E DISCUSSÃOA rota de síntese para a obtenção dos produtos planejados é mostrada na Figura 1. A obtenção dos derivados acilidrazônicos iniciou-se com a preparação do intermediário hidrazida, preparado em duas etapas a partir da esterificação do ácido 4-aminobenzoico, seguido da hidrazinólise desse éster com hidrato de hidrazina. As reações que levaram à obtenção dessa hidrazida compreenderam a sequência sintética mais comumente empregada nesse sentido. Os rendimentos obtidos para os intermediários 1, 2 e 3 foram ligeiramente inferiores aos relatados, todavia obtidos em quantidade e pureza suficientes para as reações seguintes pretendidas.A síntese de hidrazonas e acilidrazonas é feita, classicamente, por meio da condensação de aldeídos com hidrazinas e hidrazidas, respectivamente.14 O emprego de catálise ácida (ácido clorídrico, acético ou 4-toluenossulfônico, por exemplo) é relatado nas reações à temperatura ambiente ou, na ausência desses, reações em aquecimento sob refluxo.15 Essa reação se desenvolve bem em uma ampla variedade de solventes orgânicos e foi também relatada a obtenção de hidrazonas em meio aquoso ou na ausência de solventes, condições estas favorecidas pelo uso de irradiação ultrassônica ou de
