The objective of the present study was to prepare and characterized de-esterified tragacanth-chitosan nanoparticles (DET-CS NPs) as a novel carrier for methotrexate, with a view to improve drug efficacy and target ability. The preparation process was optimized using Taguchi design. NPs were characterized for size, zeta potential, morphology, thermal stability, loading efficiency, cytotoxicity and cellular uptake. Taguchi design indicated that the molecular weight of chitosan possessed the most effect on the zeta potential, PDI, and zeta deviation, and the size of nanoparticles was significantly affected by the DET concentration. The size and zeta potential of drug loaded nanoparticles at optimum condition were 322.9 ± 26 nm and 17.3 ± 5.73 mV, and thermal analysis indicated ionic bond between DET and CS in NPs. The loading efficiency was 20.32% ± 2.01, and the sustained release was observed within nine days. The IC was 280 µg/mL in HT-29, and the mitochondrial membrane potential in HT-29 was reduced more than that in MCF-7. The uptake of NPs in HT-29 was higher than that in MCF-7, and active endocytosis was the key mechanism of uptake. These phenomena altogether make DET-CS NPs a proper choice for targeted drug delivery to cells containing asialoglycoprotein receptors.

Three novel Tl(III) complexes (C1), (C2) and (C3) were synthesized using the one-pot reactions of pyridine dicarboxylic acid derivatives, 2-aminobenzimidazole and/or 4-aminopyridine, and also thallium(III) nitrate trihydrate metal salt. The structure of all three complexes was determined by the single-crystal X-ray diffraction. C1 and C2 were realized to be isostructural with disordered square anti-prismatic geometry and for C3 arrangement of the distorted tricapped triangular prism was proposed. Cyclic voltammetry measurements on the complexes exhibited that formal potential values are more positive for C1 (E0ˊ 0.109 V) and C3 (E0ˊ 0.244 V) compared to C2 (E0ˊ –0.051 V), versus Ag/AgCl under argon. Moreover, cytotoxicity of the compounds was evaluated in vitro against two cancer cell lines including a human melanoma (A375), a human colon adenocarcinoma (HT29), and also one normal cell human foreskin fibroblast (HFF). The selective and potent cytotoxicity effect was exhibited by C1 and C3 on cancer cell lines. The apoptosis through a caspase-dependent mitochondrion pathway was confirmed by ROS production, MMP reduction, p53 activation, Bax up-regulation, and Bcl-2 down-regulation, cytochrome c release, procaspase-9, and 3 expression, for A375 cells treated to C1 and C3. According to similar cellular uptake of the complexes in A375 cell line, the generation of ROS was considered as an effective agent to justify the inhibition effect C1 and C3 on mentioned cells. Furthermore, arresting the cell cycle in the G2-M phase and inducing apoptosis were indicated by these two complexes.

Background: Prosopis fracta is a valuable and useful medicinal plant for the purpose of extracting flavonoids and it is one of the most important plants with medicinal properties. Prosopis fracta is found in many southern regions of Iran. Objective: the aim of this review Phytochemistry and biological activity of Prosopis fracta. Material and Methods: Initially, the powder was degreased by hexane and then after completion of the extraction process, we obtained a rotary solvent removal and the rest of the extract was frozen for retention, then it stored at -23°C. The freeze-dried samples are dissolved in methanol and permanently filtered through a 0.35 syringe filter. Results: Prosopis fracta extracts using to treat cancer, which destroying activity via mitochondrial changes, as showed a good example for increased reactive oxygen species (ROS) levels, SIRT3 activity and cell death in HT-29 related to the intestines and rectum cancer cells. Conclusion: Antimicrobial and phytochemical studies provide valuable insight into understanding media. Sometimes the idea of the route responsible for the activity of these phytochemicals against oxidizing compounds that act as a source of harmful chemical to the body is removal from service of protein and loss of function.
