Supersensitive Detection of the Norovirus Immunoplasmon by 3D Total Internal Reflection Scattering Defocus Microscopy with Wavelength-Dependent Transmission Grating

Abstract: Norovirus (NoV) is a major foodborne pathogen, and even low levels of virus can cause infection and gastroenteritis. We developed a supersensitive NoV sensor that detects NoV group-I capsid protein (NoVP) via three-dimensional (3D) total internal reflection scattering defocus microscopy (TIRSDM) with wavelength-dependent transmission grating (TG). The combination of evanescent wave scattering and TG significantly enhanced the detection sensitivity and selectivity of NoVP in first-order spectral images (n = +1)… Show more

“… 106 Moreover, 3D total internal reflection scattering defocus microscopy with wavelength-dependent transmission grating increases the spectral resolution and detects viral capsids at 820 μM. 107 In addition, the digital microfluidics platform based on electrowetting-on-dielectric separates antibody-bound microbeads for immunoassay. 108 The microfluidic method, using a monolithic disposable cartridge, facilitates hands-free saliva analysis by integrating sample pretreatment with electrophoretic immunoassays to measure viral antigens.…”

“…Han et al [170] also developed a naked eye microfluidic 3D slip paper-based analytical device for the detection of GII.4 NoV from human feces with a LOD of 9.5 × 104 gc/mL. Furthermore, Lee et al [171] developed a supersensitive sensor using a 3D total internal reflection scattering defocus microscopy, with wavelength-dependent transmission grating for the detection of NoV with a LOD of 820 yM. This method was stated to be applicable for the early stage infection of HuNoV.…”
