Herein, we investigated whether L-ascorbic acid and α-tocopherol supplementation has potential to alleviate arsenic induced immunotoxicities in thymus, spleen and circulating leukocytes. Forty-eight adult male Wistar rats were randomly divided into four groups before the treatments. Group-I (control); Group-II (sodium arsenite, 3mg/Kg/day/rat); Group-III [sodium arsenite + L-Ascorbic acid(L-AA) (200mg/Kg/day/rat) and α-tocopherol (α-T) (400mg/Kg/day/rat)]; Group-IV (L-AA and α-T). The result showed that sodium arsenite exposure (consecutive 30 days) caused weight reduction, structural alterations of thymus and spleen, accompanied by decrease in thymocyte and splenocyte counts. Decreased superoxide dismutase and catalase activities, increased malondialdehyde and protein-carbonyl contents, reduced Nrf2 and Bcl2 expression and increased p-ERK, NF-β, Bax, and cleaved-caspase-3 expression were also observed in thymus and spleen of arsenic exposed rat. Enhanced plasma ACTH and corticosterone, ROS induced apoptosis of lymphocytes were also observed. L-AA and α-T supplement has the potential to abrogate the deleterious impact of arsenic on thymus, spleen and circulating lymphocytes. Whole transcriptome analysis of leukocytes revealed that arsenic treatment augmented the expression of Itga4, Itgam, and MMP9 genes, which might help in transient migration of leukocytes through the endothelial cell layer. Supplementation with L-AA and α-T maintained Itga4, Itgam, and MMP9 gene expression within leukocytes at lower level.

Autism spectrum disorder (ASD) is a complex behavioral disorder diagnosed by social interaction difficulties, restricted verbal communication, and repetitive behaviors. Fecal microbiota transplantation (FMT) is a safe and efficient strategy to adjust gut microbiota dysbiosis and improve ASD-related behavioral symptoms, but its regulatory mechanism is unknown. The impact of the microbiota and its functions on ASD development is urgently being investigated to develop new therapeutic strategies for ASD. We reconstituted the gut microbiota of a valproic acid (VPA)-induced autism mouse model through FMT and found that ASD is in part driven by specific gut dysbiosis and metabolite changes that are involved in the signaling of serotonergic synapse and glutamatergic synapse pathways, which might be associated with behavioral changes. Further analysis of the microbiota showed a profound decrease in the genera Bacteroides and Odoribacter, both of which likely contributed to the regulation of serotonergic and glutamatergic synapse metabolism in mice. The engraftment of Turicibacter and Alistipes was also positively correlated with the improvement in behavior after FMT. Our results suggested that successful transfer of the gut microbiota from healthy donors to ASD mice was sufficient to improve ASD-related behaviors. Modulation of gut dysbiosis by FMT could be an effective approach to improve ASD-related behaviors in patients.

Both the gut microbiome and their host participate in arsenic (As) biotransformation, while their exact roles and mechanisms in vivo remain unclear and unquantified. In this study, as3mt −/− zebrafish were treated with tetracycline (TET, 100 mg/ L) and arsenite (iAs III ) exposure for 30 days and treated with probiotic Lactobacillus rhamnosus GG (LGG, 1 × 10 8 cfu/g) and iAs III exposure for 15 days, respectively. Structural equation modeling analysis revealed that the contribution rates of the intestinal microbiome to the total arsenic (tAs) and inorganic As (iAs) metabolism approached 44.0 and 18.4%, respectively. Compared with wild-type, in as3mt −/− zebrafish, microbial richness and structure were more significantly correlated with tAs and iAs, and more differential microbes and microbial metabolic pathways significantly correlated with arsenic metabolites (P < 0.05). LGG supplement influenced the microbial communities, significantly up-regulated the expressions of genes related to As biotransformation (gss and gst) in the liver, down-regulated the expressions of oxidative stress genes (sod1, sod2, and cat) in the intestine, and increased arsenobetaine concentration (P < 0.05). Therefore, gut microbiome promotes As transformation and relieves As accumulation, playing more active roles under iAs stress when the host lacks key arsenic detoxification enzymes. LGG can promote As biotransformation and relieve oxidative stress under As exposure.
