Background and purpose: Beside being rich with sweet glycosides, Stevia rebaudiana Bertoni is an interesting source of flavonoids and phenols. The current study aimed to assess the potential for increasing total phenols and flavonoids in S. rebaudiana tissue by varying concentrations of plant growth regulators (PGR) and pH levels of media. Materials and methods:The culture was established from seeds and propagated shoots were cultured on media of different pH levels (4.6, 5.8 and 7.4) and PGR. Total polyphenolics and free radical scavenging capability of leaf, callus and root extracts were determined. Shoot height, root length, shoot and root number were also recorded. Results: Shoot elongation and root development was stimulated by singly applied growth regulators though the values of both parameters were also satisfactory in PGR-free media. In the latter media, pH value of 4.6 was a main factor for increasing leaf metabolite levels. A most significant rise in phenols and flavonoids was evident in response to combination of BA either with GA3 or IAA compared to singly applied regulators indicating synergistic effects of PGR (especially of auxins and cytokinins). However, polyphenolics levels and their distribution between different tissues were also influenced by medium pH value. A positive correlation was found between antioxidant activity of S. rebaudiana extracts and phenols and flavonoids. Conclusions: The results show that PGR and medium pH value strongly affect accumulation of secondary metabolites and can lead to significant enhancement in productivity of bioactive polyphenolics in S. rebaudiana plant cultures. INTRODUCTION S tevia rebaudianaBertoni is a small perennial shrub of the Asteraceae family, native to South and Central America. S. rebaudiana occurs naturally on infertile, acid (pH 4 to 5) soils, but thrives with soil pH as high as 7.5 [1,2]. The plant is known worldwide principally for its high content of sweet diterpene glycosides, rebaudioside A and stevioside (the most prevalent glycoside) in leaf tissue and is being used as a low-calorie and non-toxic sweetener. Moreover, antimicrobial, antiviral, anticarcinogenic and antioxidant activity of S. rebaudiana leaf extracts was demonstrated by a number of studies. Wzbudziła zainteresowanie żywieniowców dzięki dwóm związkom: rebaudiozydowi A i stewiozydowi. Wymienione glikozydy mają właściwości słodzące do 300 razy silniejsze od sacharozy. Roślina charakteryzuje się rów-nież wysoką aktywnością przeciwutleniającą wynikającą m.in. z dużej zawartości polifenoli. Ekstrakty liści stewii wykazują korzystne działanie prozdrowotne. Wiąże się to ze specyficznym działaniem rośliny, obejmującym właściwości: przeciwzapalne, hipoglikemiczne, hipotensyjne, modulujące procesy immunologiczne, przeciwpróchnicze i przeciwwirusowe.Słowa kluczowe: Stevia rebaudiana, stewiozyd, słodzik niskokaloryczny, właściwości prozdrowotne Wprowadzenie Nadmierna konsumpcja cukru może być przyczyną problemów zdrowotnych, dlatego wciąż poszukuje się alternatywnych substancji o dużej mocy słodzącej, a równo-cześnie o małej kaloryczności. Najbardziej znanymi dodatkami do żywności, umożli-wiającymi wyeliminowanie sacharozy, są substancje intensywnie słodzące. Większość z nich należy do związków syntetycznych. Ich często metaliczny posmak nie zapewnia korzystnych wrażeń smakowych, a stosowanie niektórych związków może powodować uboczne skutki zdrowotne.Stevia rebaudiana, będąca źródłem naturalnych związków słodzących, może być pożądanym zamiennikiem cukru. The aim of this research was to investigate and compare the antioxidant, anti-tyrosinase, anti-aging, and antiinflammatory activities of 16 herbal extracts for topical application in cosmetic/cosmeceutical products. Herbal plant materials were extracted by infusion in boiled water for 15 min. The total phenolic content and total flavonoid content of each extract were investigated by the Folin-Ciocalteu and aluminum chloride methods, respectively. Antioxidant activities were investigated using 2,2'-diphenyl-1-picrylhydrazyl and a ferric reducing antioxidant power assay. Anti-tyrosinase and anti-aging activities were investigated using an in vitro enzymaticspectrophotometric method. Anti-inflammatory activities were investigated using an enzyme-linked immunosorbent assay. The findings show that the Stevia rebaudiana extract has the most significant levels of both phenols and flavonoids (p < 0.05). The S. rebaudiana, Rosa damascene, and Phyllanthus emblica extracts possessed the most significant antioxidant activities (p < 0.05) and a promising whitening effect with moderate antityrosinase activities. Furthermore, the Echinacea purpurea extract possessed the most significant anti-collagenase (78.5 � 0.0 %), anti-elastase (69.0 � 1.4 %), and anti-hyaluronidase activity (64.2 � 0.3 %). The Morus alba extract possessed the most significant anti-inflammatory activity since it could inhibit the secretion of interleukin-6 and tumor necrosis factor-α (p < 0.05). Therefore, these herbal extracts have promising skin benefits and have potential for use as active ingredients in cosmetic/cosmeceutical products.
