Structure-Activity Relationship of Cinnamaldehyde Analogs as Inhibitors of AI-2 Based Quorum Sensing and Their Effect on Virulence of Vibrio spp

Abstract: BackgroundMany bacteria, including Vibrio spp., regulate virulence gene expression in a cell-density dependent way through a communication process termed quorum sensing (QS). Hence, interfering with QS could be a valuable novel antipathogenic strategy. Cinnamaldehyde has previously been shown to inhibit QS-regulated virulence by decreasing the DNA-binding ability of the QS response regulator LuxR. However, little is known about the structure-activity relationship of cinnamaldehyde analogs.Methodology/Principal… Show more

“…Cinnamaldehyde inhibited phosphorylation of phospholipase Cγ1 (PLCγ1) [17] and ERK1/2 [18]. Not only cinnamaldehyde, also its derivatives have diverse biological activities [35][36][37][38][39][40]. In particular, phydroxycinnamic acid, 4-hydroxy-3-methoxycinnamaldehyde and α-bromo-4-chlorocinnamaldehyde have the immunomodulatory effects in mammalian cells [36,37,39].…”

“…The microtiterplate was then incubated for 24 h at 30°C and fluorescence from GFP expression was measured at λ ex 475 nm and λ em 515 nm, using a Victor Wallac 2 multilabel counter (Perkin Elmer). The protease activity assay was conducted as previously described [34].…”

“…Several natural molecules have also been identified as potential AI-2-signalling inhibitors (reviewed in , including brominated furanones extracted from the algae Delisea pulchra (Ren et al, 2001(Ren et al, , 2004a and cinnamaldehyde, although the latter also appears to be a more general inhibitor of Vibrio quorum sensing through inhibition of LuxR binding to DNA (Niu et al, 2006;Brackman et al, 2011).…”
