Fucoidan is a sulphated polysaccharide isolated from brown seaweeds. It has attracted a lot of attention due to its multiple physiological and biological properties. The most dominant polysaccharides in brown seaweeds are laminarin and alginic acid. Fucoidan extracted from brown seaweed possess various biological functions including anti‐inflammatory, immunomodulatory, antitumour, antibacterial, antiviral, anticoagulant, antioxidant, neuroprotective, cardio‐protection and growth‐promoting effects. Dietary supplementation of fucoidan showed significant therapeutic influences on aquatic organisms, human, livestock and poultry. This review highlights the medicinal importance and nutritional values of fucoidan polysaccharides for fish, animals and humans, to be used as a natural agent for the treatment of various pathological disorders and to replace the synthetic antibiotics. Previously, most studies are focused on in vitro model and in mice. Further studies are warranted on in vivo model and focusing on a molecular basis to understand the mechanisms of action of fucoidan as an alternative therapeutic drug for fish, animals and humans. Another objective of this review is to give more light about the beneficial impacts of fucoidan on productive performance of animals and poultry as well as aquatic organisms.

This work reports the detailed structure of fucoidan from Sargassum miticum (2SmF2) and its ability to potentiate the inhibitory effect of glycolysis inhibitor 2-deoxy-d-glucose (2-DG). 2SmF2 was shown to be sulfated and acetylated galactofucan containing a main chain of alternating residues of 1,3- and 1,4-linked α-l-fucopyranose, fucose fragments with monotonous 1,3- and 1,4-type linkages (DP up to 3), α-d-Gal-(1→3)-α-L-Fuc disaccharides, and 1,3,4- and 1,2,4-linked fucose branching points. The sulfate groups were found at positions 2 and 4 of fucose and galactose residues. 2SmF2 (up to 800 µg/mL) and 2-DG (up to 8 mM) were not cytotoxic against MDA-MB-231 and SK-MEL-28 as determined by MTS assay. In the soft agar-based model of cancer cell colony formation, fucoidan exhibited weak inhibitory activity at the concentration of 400 µg/mL. However, in combination with low non-cytotoxic concentrations of 2-DG (0.5 or 2 mM), 2SmF2 could effectively inhibit the colony formation of SK-MEL-28 and MDA-MB-231 cells and decreased the number of colonies by more than 50% compared to control at the concentration of 200 µg/mL. Our findings reveal the metabolically oriented effect of fucoidan in combination with a glycolysis inhibitor that may be beneficial for a therapy for aggressive cancers.

Fucoidan is one of the main polysaccharides of brown algae and echinoderm, which has nutritional and pharmacological functions. Due to the low molecular weight and exposure of more sulfate groups, oligo-fucoidan or fucoidan oligosaccharides have potential for broader applications. In this research, a novel endo-α-1,4-L-fucoidanase OUC-FaFcn1 which can degrade fucoidan into oligo-fucoidan was discovered from the fucoidan-digesting strain Flavobacterium algicola 12,076. OUC-FaFcn1 belongs to glycoside hydrolases (GH) family 107 and shows highest activity at 40 °C and pH 9.0. It can degrade the α-1,4 glycosidic bond, instead of α-1,3 glycosidic bond, of the fucoidan with a random tangent way to generate the principal product of disaccharide, which accounts for 49.4% of the total products. Therefore, OUC-FaFcn1 is a promising bio-catalyst for the preparation of fucoidan-derived disaccharide. These results further enrich the resource library of fucoidanase and provide the basis for the directional preparation of fucoidan-derived oligosaccharide with specific polymerization.
