RESUMOA mastite é uma infecção frequente da glândula mamária, sendo a principal doença que afeta o úbere das fêmeas e responsável pelos maiores prejuízos à produção leiteira em caprinos, reduzindo em quantidade e depreciando a qualidade do leite e seus derivados. Nesse estudo foram colhidas amostras de leite de cabras com reação positiva ao CMT, em propriedades localizadas em Bonfim de Feira, Barra do Pojuca, Feira de Santana, Camaçari, Anguera e Antônio Cardoso, todas próximas à Cidade de Salvador, Bahia. Do material coletado foi realizado cultivo bacteriológico em ágar sangue de ovino a 10%, ágar McConkey e também enriquecido em caldo BHI. O Staphylococcus spp. foi a bactéria de maior prevalência (90,3%), seguida de Streptococcus (4,8%), Micrococcus (1,6%) e Enterobacter (1,6%). Todas as bactérias foram submetidas ao teste de sensibilidade aos antimicrobianos pelo método de difusão em disco, sendo que 54,1% das estirpes estudadas apresentaram sensibilidade a todas as drogas testadas, e 45,9% das estirpes identificadas apresentaram resistência a pelo menos um dos antimicrobianos. O presente estudo confirmou o Staphylococcus spp. como a principal bactéria envolvida na etiologia das mastites subclínicas caprinas. ABSTRACT BACTERIA INVOLVED IN SUBCLINICAL MASTITIS IN GOATS IN THE REGION OF SALVADOR, BAHIA, BRAZIL.Mastitis is a common infection of the mammary gland, the main disease affecting the udder of females, and responsible for major losses in milk production in goats, decreasing the quantity and quality of the milk and its derivatives. In this study milk samples were collected from goats with positive reaction to CMT in properties located in Bonfim de Feira, Barra do Pojuca, Feira de Santana, Camaçari, Anguera and Antônio Cardoso, all of which are places surrounding the city of Salvador, Bahia, Brazil. The samples were submitted to bacteriological culture carried out in agar with 10% sheep blood, McConkey agar, and agar enriched with BHI broth. Staphylococcus spp. bacteria was the most prevalent (90.3%), followed by Streptococcus (4.8%), Micrococcus (1.6%) and Enterobacter (1.6%). All bacteria were subjected to antimicrobial susceptibility testing by the disc diffusion method, revealing that 54.1% of the strains studied were sensitive to all drugs tested, while 45.9% of identified strains showed resistance to at least one of antimicrobials. The present study confirmed Staphylococcus spp. as the main bacteria involved in the etiology of subclinical goat mastitis.

The aim of this study was to determine the mechanisms of staphylococcal resistance to methicillin. CNS (n=100 isolates) were prepared from the mammary inflammatory secretions of 86 cows from farms located in the Lublin region.Methicillin-resistant isolates constituted 20.0% of all CNS. Staphylococcus sciuri (n=8) and Staphylococcus xylosus (n=6) were most abundant, followed by Staphylococcus chromogenes (n=3), Staphylococcus haemolyticus (n=2) and Staphylococcus warneri (n=1). The mecA gene was found in 50.0% of MRCNS (10.0% of all CNS isolates) belonging to two species: S. sciuri and S. xylosus. All mecA-positive isolates contained the protein of low affinity to penicillin (penicillin-binding protein 2a -PBP2a). The enzyme hydrolysing the β-lactam ring in antibiotics was detected in 40.0% of MRCNS; 10.0% of MRCNS isolates were characterised by the presence of the mecA gene and ability to produce β-lactamase. The remaining 20.0% of MRCNS isolates showing phenotypic resistance to methicillin were mecA gene-negative and were not able to produce β-lactamase.

Bap-dependent biofilm formation by pathogenic species of Staphylococcus: evidence of horizontal gene transfer?

The biofilm-associated protein (Bap) is a surface protein implicated in biofilm formation by Staphylococcus aureus isolated from chronic mastitis infections. The bap gene is carried in a putative composite transposon inserted in SaPIbov2, a mobile staphylococcal pathogenicity island. In this study, bap orthologue genes from several staphylococcal species, including Staphylococcus epidermidis, Staphylococcus chromogenes, Staphylococcus xylosus, Staphylococcus simulans and Staphylococcus hyicus, were identified, cloned and sequenced. Sequence analysis comparison of the bap gene from these species revealed a very high sequence similarity, suggesting the horizontal gene transfer of SaPIbov2 amongst them. However, sequence analyses of the flanking region revealed that the bap gene of these species was not contained in the SaPIbov2 pathogenicity island. Although they did not contain the icaADBC operon, all the coagulase-negative staphylococcal isolates harbouring bap were strong biofilm producers. Disruption of the bap gene in S. epidermidis abolished its capacity to form a biofilm, whereas heterologous complementation of a biofilm-negative strain of S. aureus with the Bap protein from S. epidermidis bestowed the capacity to form a biofilm on a polystyrene surface. Altogether, these results demonstrate that Bap orthologues from coagulase-negative staphylococci induce an alternative mechanism of biofilm formation that is independent of the PIA/PNAG exopolysaccharide.
