Chitosan beads loaded with ciprofloxacin hydrochloride were fabricated by ionic cross-linking with sodium tripolyphosphate. The beads showed an excellent water retention property. The degradation of fabricated beads was influenced by the pH of test medium. High drug load was achieved within the bead with a short curing time. Drug release was high in acidic medium (pH 1.2) vis-à-vis intestinal medium (pH 7.4). Ciprofloxacin hydrochloride release increased with an increasing concentration of ciprofloxacin and decreasing proportion of chitosan. Drug release followed both first-order and Higuchi's root time kinetics showing non-Fickian release mechanism.

Preparation of dexamethasone (Dex)-loaded chitosan beads were developed applying a surface-response methodology. The operational conditions were optimized with the aim of maximizing the loading capacity of Dex. The loading of the drug within the chitosan beads was carried out by ionotropic gelation technique with tetra sodium phosphate (TSP) as cross-linking agent with co-axial method that compared with simple mixing method. The co-axial method included loading of drug during dripping process using two separate syringes, simultaneously. The highest drug loading capacity for beads prepared by mixing and coaxial method was obtained 53% and 98% respectively. The beads were evaluated for surface and bulk morphology using SEM. The beads produced from mixing showed a good spherical shape with a smooth surface but co-axial method led to form a wrinkle surface. Bulk structure of Dex-loaded beads in both preparation methods had several crack regions. The optimized drug-loaded chitosan beads prepared by co-axial method revealed during drug release in the range of 40 to 80 days and percent cumulative drug released was between 25% and 50%. Drug release kinetic study was investigated via fitting with two diffusion-based semiempirical models. Also, the rate constants and diffusion exponent for the drug release were calculated. The drug release was predicted to be Fickian and/ or quasi-Fickian diffusion.

S albutamol sulfate microcapsules with a coat consisting of sodium alginate and mucoadhesive polymer such as sodium carboxy methyl cellulose (NaCMC), methyl cellulose (MC), carbopol-934, and hydroxy propyl methyl cellulose (HPMC) were prepared by ionotropic gelation technique and were evaluated for morphological characters, drug content, loading efficiency, drug-polymer interactions, swelling ratio, mucoadhesive properties, and in vitro release. The resulting microcapsules were discrete, spherical, and free-flowing, and microencapsulation efficiency was 51.28-96.70%. The microcapsules prepared with alginate alone (A4) have exhibited good mucoadhesive property in the in vitro washoff test. The swelling ratio of microcapsules was enhanced with increased alginate concentration. Salbutamol sulfate release from these mucoadhesive microcapsules was slow and extended over a period of 8 h and depends upon the concentration of the alginate. The drug release from alginate-HPMC/carbopol microcapsules followed diffusion-controlled first-order kinetics. The release rate of alginate-HPMC microcapsules (A4H) was higher than other formulations and comparable with commercially available controlled-release capsules. Microcapsules with alginate alone (A4) followed diffusion mechanism. In conclusion, alginate-HPMC/carbopol mucoadhesive microcapsules could be promising vehicle for oral controlled release of salbutamol sulfate.
