Soluble P-selectin as Parameter for Platelet Activation during Storage

Abstract: SummaryPlatelet concentrates stored at room temperature deteriorate. The so-called storage lesion is characterised by morphological changes and a loss of functionality. To find an assay for early platelet activation in platelet concentrates the morphological score, β-TG release and P-selectin expression were determined, and compared with the amount of soluble P-selectin. An ELISA was used to quantify soluble P-selectin in the storage medium. We found a significant correlation between the amount of soluble P-se… Show more

“…There can be 3 potential explanations for this. First, other than active cleavage or shedding of platelet membrane-bound P-selectin, there is evidence that sP-selectin may be released from the platelets by "direct" secretion (36,37) or passive diffusion (41). Moreover, some contribution from the endothelium is possible, although remote (42)(43)(44).…”

“…In vitro studies have shown that the soluble P-selectin ELISA is more sensitive than flow-cytometric analysis of the percentage of P-selectin-positive cells and allows earlier detection of platelet activation. 14 Statistical analysis. Analysis of paired data was performed by comparing the S1 and S2 samples with S4 from the same patient by 2-tailed Student's t-test.…”

“…For example, the endothelial cell stimulant DDAVP (desmopressin) induces raised levels of von Willebrand factor but not soluble P-selectin in vivo (Jilma et al, 1996). Furthermore, we have been unable to detect P-selectin, but plentiful von Willebrand factor, by enzyme-linked immunosorbent assay (ELISA), in human umbilical vein endothelial cell lysates or tissue culture supernatants (data not shown), whereas P-selectin is easily detected in platelet lysates and from the 'supernatants' of aged endothelialcell-free platelet concentrates designed for transfusion (Kostelijk et al, 1996). However, more pertinent data arise from the measurement of soluble P-selectin in paired serum and plasma.…”
