Restriction fragment length polymorphisms (RFLPs) were described for the porcine loci for 0-glucosidase (GBA) and the p-polypeptide 1 of the Na+,K+-transporting ATPase (ATPlBl). Linkage analyses using a three-generation pedigree provided evidence for the assignment of ATPlBl, GBA and two microsatellite loci (SO001 and S0067) to a previously described linkage group comprising the loci for blood group L (EAL) and an anonymous microsatellite (s0097). The linear order of the six markers was determined with confidence by multipoint analyses and the length of the linkage group was estimated at 88 CM. This linkage group was assigned to pig chromosome 4 on the basis of a previous physical localization of the ATPlBl gene. In situ hybridization data for SO001 presented in this study were consistent with a localization on chromosome 4 and suggested a regional localization to 4p12-pl3. The present study reveals conflicting data concerning the genetic localization of the K88 loci controlling the expression of the receptors for the E. coli pilus antigens. One group has reported data suggesting a loose linkage between K88 and EAL, now mapped to chromosome 4, whereas two other groups have found linkage between K88 and the transferrin locus (TF), mapped to chromosome 13 by in situ hybridization.

Suspensions of highly viable (< 95 To) granulocytes minimally contaminated by other cell types were isolated from the peripheral blood of pigs by a single centrifugation with low molecular weight dextran and after preferential lysis of erythrocytes by hypotonic shock. A complement-dependent cytotoxic test showed the presence of antigens of the SLA major histocompatibility complex, the SLB leucocyte system and the A and E blood group systems on the granulocytes. Some SLA typing reagents against class I (SD) antigens did not react with granulocytes, however, or yielded dubious reactions. The findings showed that the reactivity of SLA sera resembles the reactivity of human HLA sera. The results also show that compatibility in the SLA, SLB, A and E systems will have to be taken into account when preparing alloirnrnune sera for the determination of granulocyte-specific antigens of pigs.

A new, non-MHC cell membrane leucocyte alloantigen was detected in pigs by the complement dependent lymphocytotoxic technique. The new leucocyte system was designated SLD. Its product antigen SLD-1 was demonstrated to segregate independently of the SLA, SLB, SLC, A and E antigens. Family studies supplied evidence of a dominant inheritance of SLD-l. Since an allelic antigen could not be demonstrated, only two alleles for this locus are reported, namely SLDl and SLD-. No evidence of linkage was detected between the above mentioned leucocyte alloantigenic systems and SLD. The antigen was detected on enriched suspensions of T and B cells from peripheral blood, but it was not detected on erythrocytes, granulocytes and thrombocytes.
