Simultaneous determination of three active components in rat plasma by UPLC–MS/MS: Application to pharmacokinetic study after oral administration of Herba Sarcandrae extract

Abstract: A rapid, specific and sensitive ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for determination of isofraxidin, rosmarinic acid and kaempferol-3-O-glucuronide in rat plasma using warfarin as an internal standard (IS). Separation was conducted on a Thermo Hypersil GOLD C column with linear gradient elution using methanol and water. Mass spectrometric detection was conducted using selected reaction monitoring (SRM) via an electrospray ionization … Show more

“…Peak 12 had a deprotonated ion at m/z 477 and a diagnostic daughter ion at m/z 301, characteristic features of quercetin-O-glucuronide. In the same manner, peak 15 showed the characteristic fragmentation pattern of kaempferol-O-glucuronide [33]. The isomer compounds at peaks 13 and 14 showed deprotonated ions at m/z 447 but different daughter ions at m/z 285 and 301, characteristic of kaempferol-O-glucoside [34] and quercetin-O-rhamnose [35], respectively.…”

“…It was found that K3O compounds exist in many kinds of Chinese herbs, offering different levels of efficacy, and in the pharmacokinetic study of related Chinese herbs, K3O had a higher blood concentration and longer half-life. This means that K3O may be more likely to enter the body through the intestinal barrier to exert its therapeutic effect [ 38 , 39 ]. Therefore, pharmacokinetic studies of K3O will be a potential direction in further research.…”

“…Chromatographic separation was achieved on a Accucore C30 column (100 mm × 3.0 mm i.d., 2.6 μm; Thermo Fisher Scientific Inc., Waltham, MA, USA) maintained at 30 °C. A ternary solvent system, comprising water as solvent A, methanol as solvent B, and MTBE as solvent C, was used under gradient mode according to the method of Zheng et al (2017), with some modifications [ 45 ]. The initial mobile phase (0.35 mL·min −1 ) was 90% B and 5% C, increased to 95% B (in 12 min), changed to 89% B and 11% C (over 13 min), to 75% B and 25% C (over 15 min), to 50% B and 50% C (in 20 min), followed by a reconditioning at initial conditions for 2 min.…”
