Simultaneous determination of noradrenaline and dopamine in Portulaca oleracea L. by capillary zone electrophoresis

Abstract: A simple and rapid capillary electrophoretic method was developed for the simultaneous determination of noradrenaline (NA) and dopamine (DA) in Portulaca oleracea L. The buffer solution used in this method was 40 mM tris (hydroxymethyl) aminomethane (Tris)-H3PO4 at pH 2.00 containing 15% methanol. The effects of pH value, organic modifier, and applied voltage were investigated. The linear ranges of NA and DA were 0.5-100 microg/mL (r=0.9952) and 6.25-200 microg/mL (r=0.9992), respectively. The relative standar… Show more

“…Capillary electrophoresis (CE) has the advantages of excellent separation efficiency, rapid analysis and minimal use of samples and solvents for complex samples. It is an alternative technique for the separation and detection of several species of pharmaceutical importance that are extracted and used in traditional Chinese herbal medicine (Jiang et al, 2003;Yue et al, 2004Yue et al, , 2005.…”

“…There are also valid documents for the presence of biologically active catecholamines including noradrenaline, dopamine and L-dopa in P. oleracea. Large concentrations of noradrenaline in the fresh plant have been reported as 2.5 mg/g which is probably larger than that of extractable amount from the mammalians suprarenal glands (22). These neurotransmitters may be responsible for increasing blood pressure in this model of hypertension in which dexamethasone intensifies the reactivity to vasopressor effects of catecholamines.…”

“…Generally, at equal molarities, oligoprotic buffers like citrate will give substantially higher currents than monoprotic species such as acetate. Therefore, type and concentration of the buffer for CZE should be optimized to obtain a good resolution of the interested compounds in a satisfactory time [40,42,43,48,58,63,[87][88][89][90][91][92][93][94][95][96][97][98].…”

“…Methanol, ACN, 1-propanol and 2-propanol can be used to improve the selectivity and resolution of CZE by improving both the separation and the peak shape [101,102], increasing the solubility [48,64], changing the migration time [50,91,103,104] and/or decreasing the adsorption and diffusion [88] of analytes. Another way of manipulating EOF and reducing analyte interaction with the silica wall is to add amines to the buffer.…”
