Simultaneous Detection of Dual Nucleic Acids Using a SERS-Based Lateral Flow Assay Biosensor

Abstract: A new class of surface-enhanced Raman scattering (SERS)-based lateral flow assay (LFA) biosensor has been developed for the simultaneous detection of dual DNA markers. The LFA strip in this sensor was composed of two test lines and one control line. SERS nano tags labeled with detection DNA probes were used for quantitative evaluation of dual DNA markers with high sensitivity. Target DNA, associated with Kaposi’s sarcoma-associated herpesvirus (KSHV) and bacillary angiomatosis (BA), were tested to validate the… Show more

“…Jaebum Choo's team presented a few modifications of a sensor where malachite green isothiocyanate was adsorbed on gold nanoparticles with detection DNA and then hybridized with the target DNA and capture DNA (modified with biotin and streptavidin). Two of their works have applied to lateral flow strips, where this structure has been used as an alternative to the typical oligonucleotide-labelled AuNPs [90,91]. Their sensor was able to detect two DNA markers simultaneously.…”

“…Colorimetric biosensor is a simple and intuitive method for biomolecules assay. Quantitative detection of biomarkers can be achieved by distincting the generated color signal with naked eyes or a simple absorption spectra device . A commonly used type of signal label, gold (Au) nanoparticles with inherent red color, has been widely applied to detect various biomarkers .…”
