Silicon chips detect intracellular pressure changes in living cells

Abstract: The ability to measure pressure changes inside different components of a living cell is important, because it offers an alternative way to study fundamental processes that involve cell deformation. Most current techniques such as pipette aspiration, optical interferometry or external pressure probes use either indirect measurement methods or approaches that can damage the cell membrane. Here we show that a silicon chip small enough to be internalized into a living cell can be used to detect pressure changes in… Show more

“…In cell culture, FRET has also been used to measure uniaxial compression (Paszek et al, 2014). More generally, fluorescent materials exhibit a variety of properties that could be dependent on other mechanical quantities, such as intracellular pressure (Gomez-Martinez et al, 2013;Watanabe et al, 2013), and thus could be used to develop new force probes.…”

“…The two opposite surfaces of the cantilever act as mirrors that enhance the light reflectivity for certain wavelengths in which constructive interference occurs between the multiple reflections from the two mirrors. Conversely, the reflectivity is suppressed for other wavelengths by destructive interference 31,32 . This is illustrated in the modulation of the reflectivity with the wavelength observed in the cantilever with respect to the preclamping, in which interference effects are negligible (Fig.…”

“…Because the photonic microchips are planar, the total cell volume consumed by the photonic chips is limited. Therefore, the photonic chips do not seem to have a strong influence on cell viability [34], although their diameter of a few micrometers is reasonably large when compared to typical mammalian cells.…”
