Separation of cocaine stereoisomers by capillary electrophoresis using sulfated cyclodextrins

Abstract: A capillary electrophoretic method for the separation of cocaine and its stereoisomers was developed. In this study, the effect of organic modifier was also investigated. The separation was achieved using 1% sulfated cyclodextrin, 10 mmol L(-1) phosphate buffer, 10% methanol at pH 3. The method provides good reproducibility and easy application.

“…The same sulfated ␤-cyclodextrin proved useful for separation of cocaine enantiomers and diastereomers, i.e. (-)-cocaine (=(2R,3S)-cocaine, the active enantiomer formed in coca plants), (+)-cocaine, (-)-pseudococaine, and (+)-pseudococaine [170]. Zandkarimi and colleagues [171] explored highly sulfated cyclodextrins for CE enantioseparation of different drugs including atropine.…”

“…To optimize the separation of the studied drugs by CZE, the selection of the background electrolyte was based on the previously published studies in which sodium phosphate buffers were used to analyze different drugs [9,10,13]. In our work, we used disodium phosphate and tested concentrations ranging from 50 to 100 mM.…”

“…CE is an attractive alternative due to its extreme efficiency, short analysis time and wide scope of application [9][10][11][12]. However, the application of CE for drugs of abuse has been mainly focused to the optimization of the separation step [9,10,13] and to the analysis of biological samples.…”

“…A capillary electrophoretic method for the separation of cocaine and its stereoisomers has been developed by Cabovska et al [35] based on a 10 mM phosphate buffer pH 3 containing 1% sulfated CDs and 10% methanol. The use of a negatively charged CD, having a mobility opposite to the cationic analytes, showed a higher retarding effect and, reportedly, a better resolution in comparison to native CDs.…”
