Screening for Voltage-Gated Sodium Channel Interacting Peptides

Abstract: The voltage-gated sodium channel (VGSC) interacting peptide is of special interest for both basic research and pharmaceutical purposes. In this study, we established a yeast-two-hybrid based strategy to detect the interaction(s) between neurotoxic peptide and the extracellular region of VGSC. Using a previously reported neurotoxin JZTX-III as a model molecule, we demonstrated that the interactions between JZTX-III and the extracellular regions of its target hNav1.5 are detectable and the detected interactions … Show more

“…The whole-cell patch-clamp experiment was performed to analyse the electrophysiological activities of rJZTX-III. As demonstrated in Figure 5, 1 mmol/L rJZTX-III could significantly inhibit the current of hNav1.5 expressed in HEK293 cells, which was similar to the effects observed for the natural JZTX-III [15]. This result suggests that rJZTX-III expressed in the cytoplasm of the SHuffle strain through the galactose auto-induction method retains its bio-functional activity.…”

“…Chimaeragenesis is a useful tool for the study of relationships of protein structure and function. For example, by constructing different voltage-gated channel subtypes chimaeras, chimaeragenesis already revealed some important gating properties of the less-characterized sodium channels, such as Na v 1.9 and Na v 1.8 [ 8 , 9 ]. Chimaeragenesis, by which two or more DNA modules could be stitched together, is generally achieved by in vivo [ 10 ] or/and in vitro [ 11 ] recombination or overlap-extension PCR [ 12 , 13 ].…”

“…Nevertheless, other interactions might not be detected in YTH assay because of the lack of chaperones, assembly factors, post-translational modifications, or other effects 43 . In addition, the YTH assay may produce false positives if (1) the two proteins are not expressed in the same tissue or at the same time or if (2) the prey protein has a self-activating function 43 44 .…”
