Screening and characterization of a novel alkaline lipase from Acinetobacter calcoaceticus 1-7 isolated from Bohai Bay in China for detergent formulation

Abstract: A novel alkaline lipase-producing strain 1-7 identified as Acinetobacter calcoaceticus was isolated from soil samples collected from Bohai Bay, China, using an olive oil alkaline plate, which contained olive oil as the sole carbon source. The lipase from strain 1-7 showed the maximum activity at pH 9.0 under 40 o C. One interesting feature of this enzyme is that it exhibits lipase activity over a broad range of temperatures and good stability. It is also stable at a broad range of pHs from 4.0 to 10.0 for 24 h. “…Yu et al (2007) indicated that the lipase functions were normal at pH from7.0 to 8.5or at temperature from 35 °C to 45 °C. Wang et al (2012) also suggested that the lipase activity was significantly decreased above 45°C or pH below 6.0 and above 9.0.Present data suggested that the enzyme had a strong activity at 45 °C and acidic pH below 7.0. Obviously, these results were mostly in agreement with above three investigations, thus providing support to the accuracy of the method applied in thiscurrent study.…”

“…3). Similarly, various alkaline lipases that are stable and active at high temperatures were also purified from mesophiles (Sugihara et al 1991; • C was seen for the alkaline lipase of Acinetobacter calcoaceticus 1-7 (Wang et al 2012). The present lipase can therefore work at any washing temperature and this will save electricity by washing at normal temperature and the whole process will be cost effective.…”

“…This may be due to the high concentration of triton may affect the kinetic behavior of Lipozyme RMIM. Wang 22 indicated that use of 1 of triton concentration instead of 10 would enhance some enzymes stability. Therefore concentration of 1 Triton was used in this research.…”
