HPLC with quadrupole TOFMS and chemometrics analysis for the characterization of Folium Turpiniae from different regions

Abstract: Folium Turpiniae has been used as a traditional Chinese medicine for the treatment of abscesses, fevers, gastric ulcers, and inflammations. This paper describes a strategy of combining HPLC with photodiode array detection and quadrupole TOF-MS, as well as phytochemical and chemometrics analysis for the characterization, isolation, and simultaneous quantification of the chemical constituents of Folium Turpiniae. 19 constituents were identified, namely, 11 flavonoids, seven gallic acid derivates, and quinic acid… Show more

“…Folium Turpiniae, a well-known traditional Chinese medicine derived from Turpinia arguta (Staphyleaceae), is used for the treatment of abscesses, fevers, gastric ulcers, and inflammations [84]. A strategy incorporating HPLC-PDA and quadrupole TOF-MS detection, as well as phytochemical and chemometrics analysis (and fingerprinting) for the characterization, isolation, and simultaneous quantification of the chemical constituents of Folium Turpiniae has recently been reported [84].…”

“…It is valuable but difficult to identify and assign most constituents in a complex herbal medicine. Fortunately, such is the sensitive and reliable approach developed for quantifying seven alkaloids in Fuzi decoction by triple‐quadrupole MS , and the application of quadrupole TOF MS (Q‐TOF‐MS) in identifying multiple constituents of Folium Turpiniae , HPLC–MS/MS has been proved rapid and sensitive with greater accuracy and precision for structural elucidation and multiple component quantification in complex systems . Therefore, an explanation on the whole picture of chemical component and the content of key compounds in YGS is necessary and feasible.…”

“…Fragmentation of the molecular ion of component 16 ( m/z 737.2262, 12.61 min, –3.446 ppm) led 3 major product ions at m/z 591.1711 ([M‐H‐Rha] − ), 283.0614 (the [M‐H] − ion of luteolin) and 240.0435 ([M‐H‐2Rha‐Glc‐CH 3 ‐CO] − ). By comparison with the published literature, component 16 was tentatively identified as neobudofficide (Li and others ).…”
