Identification of the Major Components of Buddleja officinalis Extract and Their Metabolites in Rat Urine by UHPLC‐LTQ‐Orbitrap

Abstract: Buddleja officinalis Maxim, one of the most popular herbal medicines in China, is widely prescribed for curing eye diseases for centuries. In this study, the major components of B. officinalis extract (BOE) and their metabolites in rat urine were detected and identified by ultra-high-pressure liquid chromatography coupled with linear ion trap-orbitrap tandem mass spectrometry (UHPLC-LTQ-Orbitrap). A total of 19 compounds, including 8 flavonoids and 11 phenylethanoid glycosides, were confirmed or tentatively id… Show more

“…Phytochemical analysis of Buddleja extracts identified polyphenols, flavonoids and phenylethanoid as major components [45, 46]. Buddleja showed antimicrobial activity against Bacillus subtilis, S. aureus, E. coli and K. pneumoniae supporting the traditional use of the plant in the treatment of eye infections [28].…”

“…However, the determination method by HPLC usually took too much time (>40 min), and was not sensitive enough for trace component analysis in the complicated extracts. Recently, a hybrid linear ion trap-Orbitrap (LTQ-Orbitrap) analytical platform has been applied to the analysis of small molecules in various traditional Chinese medicine (TCM) and biological samples [13][14][15][16][17]. It consists of a 2D ion trap coupled with an Orbitrap, and allows two different scan types to be acquired simultaneously.…”

“…Hence, the overall objective of this research was to investigate in detail the in vivo absorption and metabolism of Areca catechu L. nut extract (ACNE). As we all know, plasma and urine are recognized as ideal biological matrices in drug absorption and metabolism studies [ 21 , 22 ]. In this work, a total of 26 components in the plasma and 31 components in the urine were characterized by ultra-high-pressure liquid chromatography coupled with linear ion trap-Orbitrap tandem mass spectrometry (UHPLC-LTQ-Orbitrap).…”
