Scanning electron microscopic observations of early plaque formation in vivo on enamel specimens treated with delmopinol

Abstract: The aim of this double blind, cross-over, scanning electron microscopic (SEM) study was to compare the effect of topical application of 0.5% delmopinol HCl (aqueous solution) with placebo on early supragingival plaque formation on enamel specimens attached to a maxillary premolar. 3 subjects underwent 2x (placebo and delmopinol treatment, respectively) 7 treatment periods (1/2, 1, 2, 8, 24 hours; 3, 7 days) of undisturbed dental plaque accumulation, during which 2 ml of placebo and delmopinol HCl 0.5%, respect… Show more

“…The inhibitory effect of delmopinol on plaque formation was more pronounced compared to placebo, which is in agreement with previous studies (8)(9)(10)(11)(12)(13). Comparing baselines values after professional toothcleaning of the two test periods revealed a slightly higher Plaque Index score in the second than the first test period (P=0.016).…”

“…(32.5%). These re- suits are in agreement with the presence of coccoid-dominated plaque in subjects applying deltnopinol, as observed by light atid scanning electron tnicroscopy (6,8). On day 7 an Increase in the proportion of Streptoeoecus spp., Haemophilus spp.…”

“…Although the agents did not eliminate plaque, they selectively altered the subgingival fiora with higher percentages of gram-positive cocci and lower percentages of gram-negative rods and Forphyromonas gingivalis compared to the control atiitnals. The amino alcohol, delmopitiol hydrochloride (Decapinol'") has the potential to change the composition of the supragingival plaque (5,6,8). Microscopic evaluation of the supragingival plaque adjacent to the gingival margin showed that it was dotninated by coccoid microbial forms when delmopinol was applied or rinsed with for 14 days.…”

“…No serious adverse events were reported in this study. As in previous studies (Collaert et al 1992a, b, 1993a, b, 1994, Moran et al, 1992, Rundegren et al, 1992a, Hase et al 1995a, b, Claydon et al 1996, Klinge et al 1996, a transient anaesthetic sensation, especially located at the tip of the tongue, was the dominant adverse event reported in the delmopinol group. At the 6-month examination, however, this side-effect was reported with the same frequency also in the chlorhexidine group.…”

“…The results of the microbiological examinations in this study and previous clinical studies indicate that painting (Collaert et al, 1992b(Collaert et al, , 1993a and rinsing (Collaert et al 1992a) with delmopinol delays and interferes with dental plaque maturation. In this study, the 0.2% chlorhexidine rinsing solution showed 38% lower plaque scores than placebo.…”
