The species Elaeocarpus sphaericus (Rudraksha) is a religious, medicinally important threatened tree of India. An efficient micropropagation protocol has been developed from nodal explants of this plant species collected from north-east India for large scale production of planting material at favourable sites within the country. Best shoot initiation occurred in MS medium supplemented with 2.2μM BA+2.2μM Kn in combination. Addition of Casein Hydrolysate (CH) (100mg/L) increased the shoot number. Microshoots excised and subcultured in 2.0μM BA further enhanced growth and multiplication. The shoot cultures were maintained in this concentration for 2years with subculturing at 6weeks interval. MS medium containing 5.0μM NAA was most effective for rooting. Successfully acclimatized plants (80%) showed normal growth under suitable habitat conditions.

Passiflora foetida, uma espécie de maracujá silvestre, tem sido pesquisado devido ao seu potencial farmacêutico e sua característica agronômica de resistência ao Fusarium spp., possibilitando a sua utilização como porta-enxerto para espécies de maracujá suscetíveis a morte prematura. Assim, visando a propagação dessa espécie, a micropropagação é uma das formas mais utilizadas, sendo necessário a utilização de acessos de uma planta matriz para, por organogênese, gerar novas plântulas com o mesmo genótipo. Desta forma, este trabalho teve como objetivo estabelecer protocolo para a propagação in vitro de P. foetida pela indução de gemas por organogênese, testando-se duas concentrações de sais minerais do meio MS, sem adição de fitormônios. Para o estabelecimento in vitro foram utilizados gemas apicais e laterais obtidas a partir de plantas matrizes de P. foetida, cultivadas em casa de vegetação. Os experimentos foram realizados em delineamento inteiramente casualizado. Aos 60 dias após a inoculação dos explantes foram avaliados: número de gemas, comprimento da brotação, número de folhas, escore de coloração, escore de desenvolvimento e número de raiz. Para garantir a precisão dos resultados foram realizadas técnicas estatísticas aplicadas à análise de dados e otimização dos resultados. O desenvolvimento das plantas por organogênese apresentou valores estatisticamente similares em ambas as concentrações, podendo concluir que o meio MS com metade das concentrações de nutrientes foi o mais viável para o estabelecimento in vitro dessa espécie.

Turmeric (Curcuma longa) is an Asian native species used for ornamental, medicinal and food purposes. However, its conventional propagation method is costly and inefficient. Therefore, this study aimed to verify appropriate asepsis methods for shoots from rhizomes, evaluate concentrations of activated charcoal and sucrose, and determine appropriate salt concentrations in Murashige and Skoog (MS) medium for turmeric growth. Three independent assays were performed. First, asepsis methods were verified for explants; second was for concentrations of sucrose and activated charcoal and in the final assay salt concentrations was evaluated. In all assays, MS medium supplemented with 30 g/L of sucrose, 6.5 g/L of agar and pH adjusted to 5.8 was used. Two growth regulators were added to the culture medium: 4.44 µM of 6-benzyl-amino-purine (BAP) and 1.08 µM of α-naphthalene acetic acid (NAA). Data obtained were subjected to analysis of variance (ANOVA) at p ≤ 0.05 probability. The averages were compared by Tukey's test and polynomial regression at p ≤ 0.05. All the asepsis methods evaluated showed similar effects. The highest concentration was obtained by utilizing 60 mg/L sucrose for the most part of the evaluated characteristics; however, it is recommended that activated charcoal be used at a concentration of 4.5 g/L. For salt concentrations, averages were compared by polynomial regression at (p ≤ 0.05) probability in 50 to 60% MS medium, and resulted in longer root systems, greater numbers of shoots and more leaves. The usual dose of 3% sucrose resulted in lower development of in vitro seedlings. Salt concentrations higher than 60% were toxic to turmeric tissues and consequently compromised the root system and the aerial part of developing seedlings.
