Restorative potentiality of S-allylcysteine against diabetic nephropathy through attenuation of oxidative stress and inflammation in streptozotocin–nicotinamide-induced diabetic rats

Abstract: Thus, SAC showed adequate therapeutic effect against DN by downregulation of inflammatory factors and attenuation of oxidative stress. Histological and SEM observations also indicated that SAC treatment notably reverses renal damage and protects the kidneys from hyperglycemia-mediated oxidative damage.

“…Oxidative stress is caused by either overproduction of ROS, due to stress, environmental factors, lifestyle changes, etc., or due to inadequate cellular antioxidant levels in various biological systems. The endogenous antioxidant enzyme defense system plays a significant role in scavenging the ROS in tissue injury and in maintaining cellular integrity by their reducing properties (Sathibabu Uddandrao et al., 2018, 2019). Superoxide dismutase, catalase, and GPx form the first‐line cellular defense system present in a biological system to scavenge free radicals in oxidative damage by decomposing hydrogen peroxide and superoxide radicals (Griendling & FitzGerald, 2003).…”

“…There is a balance between the production of ROS and the endogenous cellular antioxidant system, including enzymatic and nonenzymatic, under physiological condition. However, more and more animal experiments and clinical investigations indicated that diabetes not only induced the overproduction of ROS but also might interfere the antioxidant defense system via regulating the activities of antioxidant enzyme [35]. Thus, to further prove the renoprotective effect of EVA, we explored the effect of EVA on the level of SOD, CAT, and GSH-Px, important antioxidases in the antioxidant defense system in renal damage, in vivo and in vitro .…”

“…Diabetes mellitus (DM) is characterized by persistent hyperglycemia with an inability to fully utilize glucose and maintain insulin homeostasis, which leads to long-term damage and dysfunction of the eyes, kidneys, and nervous and cardiovascular systems in particular (1). Approximately 366,000,000 individuals are expected to suffer from DM by the year 2030, and almost 592,000,000 individuals, or 1/10, are anticipated to have diabetes by 2035 (1,2). It is estimated that >40% of diabetic patients require regular, painful and expensive dialysis due to end-stage renal disease (ESRD), and the mortality rate remains high (3).…”

“…Diabetic nephropathy (DN) is an emerging clinical and public health challenge, which is also a life-threatening long-term microvascular complication of type 1 DM (T1DM) and type 2 DM (T2DM) (1,4,5). Patients with DN are generally hypertensive, which increases the risk of cardiovascular disease and mortality (6); hypertension is interconnected with the initiation and progression of DN.…”
