Replication of a Temperature Sensitive Mutant of Infectious Bovine Rhinotracheitis Virus in the Tissues of Inoculated Calves1

Abstract

Summary The RLB 106 strain is a ts mutant of IBR virus. Its replication in different organs was studied after experimental inoculation of calves by the nasal route and it was compared to that of a virulent virus. In the calves inoculated with the virulent strain, virus was recovered not only from the turbinates, but also from the trachea, the tonsils, the costo‐cervical lymph nodes and, in 1 of the 2 inoculated animals, from various areas of the lungs and the bronchial lymph nodes. In the calves inoculated with the ts mutant all these tissues were negative except for the nasal mucosa and in one calf the tonsils. The spleen, blood and genital organs failed to yield virus in either group. The brain and cerebellum were also negative in all calves; so was the cerebrospinal fluid except for a small amount of virus found in the fluid from one of the calves inoculated with the ts mutant; this positive finding may have been due to contamination of the sample or to passive transfer of virus from the sites of replication. Zusammenfassung Vermehrung einer temperaturempfindlichen Mutante des Virus der infektiösen bovinen Rhinotracheitis in experimentell infizierten Kälbern Nach experimenteller intranasaler Infektion von Kälbern mit einer temperaturempfindlichen Mutante des IBR‐Virus (Stamm RLB 106) gelang die Reisolierung des Virus lediglich aus der Nasenschleimhaut und, bei einem Kalb, aus den Tonsillen und der Zerebrospinalflüssigkeit. Im Gegensatz dazu wurde bei Kälbern, die mit virulentem IBR‐Virus infiziert worden waren, das Virus nicht nur aus der Nasenschleimhaut, sondern auch aus der Trachea, den Tonsillen, den Kostozervikallymphknoten, der Lunge und den Lungenlymphknoten isoliert. Die Ergebnisse werden diskutiert. Résumé Multiplication d'un mutant sensible à la température du virus de la rhinotrachéite infectieuse bovine chez des veaux infectés expérimentalement Après avoir infecté intranasalement des veaux avec un virus mutant IBR (souche RLB 106) sensible à la température, on a pu réisoler le virus uniquement à partir des muqueuses nasales et chez un veau, à partir des amygdales et du liquide céphalo‐rachidien. Par contre, chez des veaux infectés avec un virus IBR virulent, l'agent ne fut pas seulement isolé des muqueuses nasales mais également de la trachée, des amygdales, des ganglions costocervicaux, des poumons et des ganglions pulmonaires. On discute les résultats. Resumen Multiplicación de una mutante termolábil del virus de la rinotraqueitis bovina infecciosa en ternos infectados experimentalmente Tras la infección intranasal experimental de terneros con una mutante termolábil del virus IBR (estirpe RLB 106) solo se logró el reaislamiento del virus de la mucosa nasal y, en un ternero, de las tonsilas y el líquido cerebroespinal. En contraposición a esto, se aisló el virus en terneros infectados con virus IBR virulento no solo de la mucosa nasal, sino también de la tráquea, tonsilas, ganglio linfático costocervical, pulmón y los ganglios pulmonares. Se discuten los resultados.
